SES rescue operators worked to stabilise the vehicle and release the driver, who paramedics and a Toll Rescue Helicopter crew treated at the scene before rushing to Liverpool Hospital in a critical condition.
Police said one horse died in the crash and another was euthanised due to their injuries, while seven were taken away for treatment.
The 26-year-old male tractor driver was uninjured and underwent mandatory testing.
Several lanes of the Hume Highway were closed for some time in both directions.
Police said inquiries into the crash were ongoing.
