{"id":120266,"date":"2025-09-05T07:32:09","date_gmt":"2025-09-05T07:32:09","guid":{"rendered":"https:\/\/www.newsbeep.com\/au\/120266\/"},"modified":"2025-09-05T07:32:09","modified_gmt":"2025-09-05T07:32:09","slug":"scientists-develop-way-to-make-dmd-treatments-work-better","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/au\/120266\/","title":{"rendered":"Scientists develop way to make DMD treatments work better"},"content":{"rendered":"<p>Researchers have developed a way to enhance the efficacy of exon-skipping therapies for <a href=\"https:\/\/musculardystrophynews.com\/muscular-dystrophy-types\/what-is-duchenne-muscular-dystrophy\/\" rel=\"nofollow noopener\" target=\"_blank\">Duchenne muscular dystrophy<\/a> (DMD), and the approach showed promise in a mouse model of the disease.<\/p>\n<p>The study,\u00a0\u201c<a href=\"https:\/\/www.sciencedirect.com\/science\/article\/pii\/S2162253125002197\" rel=\"nofollow noopener\" target=\"_blank\">A Combinatorial Oligonucleotide Therapy to Improve Dystrophin Restoration and Dystrophin-Deficient Muscle Health<\/a>,\u201d was published in\u00a0Molecular Therapy Nucleic Acids.<\/p>\n<p><a href=\"https:\/\/musculardystrophynews.com\/causes\/\" rel=\"nofollow noopener\" target=\"_blank\">DMD is caused<\/a> by mutations in the DMD gene, which encodes dystrophin, a protein essential for maintaining muscle cell integrity. These mutations severely reduce or eliminate dystrophin production, leading to progressive muscle damage.<\/p>\n<p>Protein-coding genes such as DMD are composed of sections called exons. When the gene is read to make a protein, the individual exons are combined to form the full gene, similar to how words are combined to form a sentence. <a href=\"https:\/\/musculardystrophynews.com\/exon-skipping-for-duchenne-muscular-dystrophy\/\" rel=\"nofollow noopener\" target=\"_blank\">Exon-skipping therapies<\/a> aim to remove a few exons from the mutated gene, allowing cells to make a shortened but functional version of the dystrophin protein. Several exon-skipping therapies are <a href=\"https:\/\/musculardystrophynews.com\/approved-treatments-for-muscular-dystrophy\/\" rel=\"nofollow noopener\" target=\"_blank\">authorized in the U.S.<\/a> to treat DMD patients with specific mutations.<\/p>\n<p>Many exon-skipping therapies use a type of molecular technology called phosphorodiamidate morpholino oligomers, or PMOs, to exert their effects on the\u00a0DMD\u00a0gene. A major limitation of this approach is that muscle cells in DMD are often in a state of inflammation and fibrosis (scarring), which makes it harder for PMOs to get into muscle cells and exert their effects.<\/p>\n<p>  Recommended Reading<\/p>\n<p>      <img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/au\/wp-content\/uploads\/2025\/09\/BettyVertin_PartyOf9-1-150x0-c-default.jpg\" alt=\"banner for Betty Vertin's \" party=\"\" of=\"\" column=\"\"\/><\/p>\n<p>Technique appears to boost therapy\u2019s efficacy<\/p>\n<p>A team of scientists in the U.S. and Korea developed a new approach aiming to improve the delivery of PMOs targeting the\u00a0DMD\u00a0gene to DMD muscle cells. To do this, the researchers used a second PMO targeting a different gene.<\/p>\n<p>They created a new PMO designed to shut down the activity of TGFB1.\u00a0This gene encodes a signaling molecule called transforming growth factor beta (TGF-beta), which plays a key role in driving muscle inflammation and fibrosis in DMD. The researchers reasoned that if they could reduce the gene\u2019s activity, they could reduce muscle inflammation and fibrosis, which would in turn make it easier for the PMO targeting the DMD\u00a0gene to get into muscle cells.<\/p>\n<p>\u201cOur novel approach combines PMO-based exon skipping therapy with TGF-[beta] inhibition to enhance the clinical efficacy of DMD therapies,\u201d the researchers wrote.<\/p>\n<p>The researchers demonstrated this approach in a mouse model of DMD. Two PMOs led to more dystrophin protein production and better muscle cell regeneration than one PMO targeting the DMD\u00a0gene.
