This maternal-fetal interface, which forms about a week after fertilization, supports fetal growth while simultaneously maintaining the mother\u2019s health.<\/p>\n<p>Now, scientists have mapped this microscopic frontier in unprecedented detail. Their goal is to uncover the exact biological triggers behind some of the most devastating pregnancy complications. <\/p>\n<p>The researchers from the University of California, San Francisco (UCSF), published a comprehensive cell atlas of the human placenta and uterus. They tracked nearly 1.2 million cells from early pregnancy to birth. By charting how these tissues actually grow, the team discovered a brand-new maternal cell type, decoded the mechanics of fetal blood supply, and pinpointed the exact cells responsible for preeclampsia, preterm birth, and miscarriage.<\/p>\n<p>\u201cThis work gives us a much clearer picture of this critical region than ever before,\u201d said Jingjing Li, associate professor in UCSF\u2019s Department of Neurology.<\/p>\n<p>A Whole-Pregnancy Perspective<\/p>\n<p>Prior to this new study, other research groups had mapped the placenta and uterus. However, those earlier efforts covered only select stages of pregnancy. As such, each investigation could only offer snapshots, rather than a full feature film.<\/p>\n<p>\u201cThe biggest difference is we are looking at the whole time course,\u201d Jingjing Li told <a href=\"https:\/\/www.livescience.com\/health\/reproductive-health\/no-one-knows-what-they-are-researchers-discover-new-type-of-cell-thats-seen-only-during-pregnancy\" rel=\"nofollow noopener\" target=\"_blank\">Live Science<\/a>.<\/p>\n<p>The new atlas incorporates data from tissues collected between weeks 5 and 39 of pregnancy. The team achieved this sweeping view by analyzing about 1.2 million placental and uterine cells.<\/p>\n<p>Specifically, they looked at 200,000 isolated cells to pinpoint their identities. Then, they examined nearly one million cells embedded in their original locations within the tissue.<\/p>\n<p>\u00d7<\/p>\n<p>                        Thank you! One more thing&#8230;<\/p>\n<p>Please check your inbox and confirm your subscription.<\/p>\n<p>Looking at isolated cells helps identify their type, but it lacks the context of \u201cwhere these cells are located within the tissue, which cells they are near, and how they interact with one another,\u201d Roser Vento-Tormo, a group leader in cellular genomics at the Wellcome Sanger Institute, and postdoc Ana Paredes told Live Science.<\/p>\n<p>By combining these techniques, the scientists could observe precisely how genes activate, how proteins interact, and how DNA packages itself as the pregnancy progresses.<\/p>\n<p>\u201cBy examining this tissue cell by cell across pregnancy, we can begin to understand both normal development and what may go wrong,\u201d said Susan J. Fisher, professor of Obstetrics, Gynecology and Reproductive Sciences at UCSF.<\/p>\n<p>The Biological Speed Bump<\/p>\n<p>As they charted this new map, the scientists stumbled upon a surprise. They revealed a subtype of maternal cell that no one had ever described before.<\/p>\n<p>These cells do not exist in the uterus outside of pregnancy. Instead, they suddenly multiply at the start of gestation as the uterine lining morphs to cradle the embryo.<\/p>\n<p>\u201cThat was an exciting moment during the study,\u201d Jingjing Li said. \u201cWe asked around \u2014 no one knows what they are.\u201d<\/p>\n<p>The researchers found these maternal cells located exactly where fetal placental cells first enter the uterus. Readers might wonder: how do fetal cells know when to stop digging into the mother\u2019s tissue?<\/p>\n<p>It turns out this newly discovered cell type acts as a biological regulator. By sending out specific signals, the cell acts as a sort of speed bump to prevent the fetal cells from proceeding too quickly and deeply into the mother\u2019s tissue, according to Li. <\/p>\n<p>\u201cIt\u2019s at the frontline of the maternal-fetal interface,\u201d Cheng Wang, the study\u2019s first author, told Live Science.<\/p>\n<p>That\u2019s not all. The researchers found that these speed bump cells carry a cannabinoid receptor. When the scientists exposed these cells to cannabinoid molecules in the lab, the cells restricted placental invasion even further. This could mean that using cannabis during pregnancy could be very bad for a baby.<\/p>\n<p>\u201cPopulation studies have linked cannabis use during pregnancy to poorer outcomes,\u201d Wang said. \u201cThis cell type may help explain the biological basis of that association.\u201d<\/p>\n<p>While this sensitivity to cannabinoids might not explain every risk posed by cannabis use in pregnancy, it provides a vital new clue for understanding poor oxygen delivery to the fetus and low birth weight.<\/p>\n<p>The Art of a Precise Invasion<\/p>\n<p>Early in pregnancy, certain fetal cells must invade the uterus and remodel the mother\u2019s major arteries. It\u2019s a highly invasive process that literally reshapes the mother\u2019s body. However, the takeover is necessary. It ensures the placenta receives enough high-velocity blood to nourish the growing baby. The new Nature paper reveals the startling mechanics of this process.<\/p>\n<p>Maternal blood vessel cells literally undergo a programmed death \u2014 a cellular suicide known as apoptosis \u2014 to make way for the advancing fetal cells.<\/p>\n<p>Using machine learning, the researchers even built a model that successfully predicted how deeply a given fetal cell would invade the uterus based entirely on its gene activity.<\/p>\n<p>\u201cFor a healthy pregnancy, this invasion needs to be very precisely controlled,\u201d Roser Vento-Tormo and Ana Paredes said. <\/p>\n<p>And when this invasion goes awry, tragedy often strikes. If fetal cells do not penetrate deeply enough, or if they penetrate too deeply, the mother and child face severe risks.<\/p>\n<p>The Genetic Blueprints of Miscarriage<\/p>\n<p>With their completed map in hand, the researchers sought to understand the origins of pregnancy complications. They integrated genetic data from more than 10,000 patients.<\/p>\n<p>Large medical studies have long known that specific gene variants increase the risk of conditions like preeclampsia, a life-threatening disorder marked by sudden high blood pressure. But scientists did not know exactly where in the body those risky genes did their damage.<\/p>\n<p>By laying the genetic risk signals over their new cell atlas, the team found the answers. They discovered that preeclampsia risk concentrates heavily in the fetal cells attempting to invade the mother, and the maternal cells lining the blood vessels.<\/p>\n<p>This strongly suggests that preeclampsia fundamentally results from disrupted communication. The maternal and fetal cells fail to coordinate the vital remodeling of the mother\u2019s arteries.<\/p>\n<p>The researchers performed the same analysis for spontaneous preterm birth and sporadic miscarriage. They traced the genetic risk for both conditions to a highly specific population of early endometrial lining cells in the mother.<\/p>\n<p>\u201cThis will help us to know which cells are underlying those complications,\u201d Jingjing Li said. <\/p>\n<p>The Next Frontier<\/p>\n<p>This monumental atlas finally gives scientists a baseline for a healthy pregnancy.<\/p>\n<p>The researchers now plan to compare this healthy baseline against pregnancies impacted by various diseases. They intend to hunt for new treatments that can target the exact cellular miscommunications causing preeclampsia and miscarriage.<\/p>\n<p id=\"h-\">\u201cIf we include more cells, more samples, a lot of new, exciting discoveries could be made,\u201d Jingjing Li concluded. \u201cSo this is really a starting point.\u201d<\/p>\n<p>The findings were reported in the journal <a href=\"https:\/\/www.nature.com\/articles\/s41586-026-10316-x\" rel=\"nofollow noopener\" target=\"_blank\">Nature<\/a>.<\/p>\n","protected":false},"excerpt":{"rendered":"Credit: Kangaroo Care.
