What\u2019s next: The funding will support a Phase 1b trial expected to start in early 2027, evaluating safety, tolerability and pharmacokinetics alongside exploratory measures of cognition and disease progression.<\/p>\n

Why it matters: Remynd is pursuing a mechanism well outside the amyloid-targeting approaches that have dominated recent Alzheimer\u2019s drug development. REM392 is designed to repair septin filaments \u2014 part of the neuronal cytoskeleton \u2014 that become disrupted by pathological tau.<\/p>\n

The company says restoring these filaments normalizes calcium influx into neurons, potentially improving synaptic function while also affecting underlying Alzheimer\u2019s pathology. The mechanism was described in a 2024 Science paper<\/a>.<\/p>\n

The backstory: REM392 is a second-generation drug. Remynd previously tested an earlier septin modulator<\/a>, REM127, in a 14-patient Phase 2a study in mild-to-moderate Alzheimer\u2019s disease. That trial was stopped early after dose-dependent liver enzyme elevations, meaning its primary safety endpoint was not met.<\/p>\n

The small study nevertheless produced exploratory signals across several biomarkers and cognitive measures, prompting Remynd to develop REM392 with the aim of retaining septin activity while improving safety. The company began testing REM392<\/a> in healthy volunteers in 2025.<\/p>\n

What they\u2019re saying: \u201cWe are moving Alzheimer\u2019s drug development beyond the usual targets and mechanisms, because we believe that is where the larger gains are for patients,\u201d Remynd CSO Gerard Griffioen said. \u201cThe planned Phase 1b study will put this to the test in patients and generate the next body of evidence.\u201d<\/p>\n
