Scientists have since realized non-coding parts of the genome are <a href=\"https:\/\/www.sciencealert.com\/scientists-find-a-crucial-new-role-for-dna-we-once-considered-junk\" style=\"--tw-scale-x: 1; --tw-scale-y: 1; --tw-scroll-snap-strictness: proximity; --tw-ring-offset-width: 0px; --tw-ring-offset-color: #fff; --tw-ring-color: #3b82f680; --tw-ring-offset-shadow: 0 0 #0000; --tw-ring-shadow: 0 0 #0000; --tw-shadow: 0 0 #0000; --tw-shadow-colored: 0 0 #0000; letter-spacing: -0.45px;\" rel=\"nofollow noopener\" target=\"_blank\">more important<\/a> <a href=\"https:\/\/www.sciencealert.com\/the-secret-to-sea-dragons-fabulous-features-is-in-their-genes\" style=\"--tw-scale-x: 1; --tw-scale-y: 1; --tw-scroll-snap-strictness: proximity; --tw-ring-offset-width: 0px; --tw-ring-offset-color: #fff; --tw-ring-color: #3b82f680; --tw-ring-offset-shadow: 0 0 #0000; --tw-ring-shadow: 0 0 #0000; --tw-shadow: 0 0 #0000; --tw-shadow-colored: 0 0 #0000; letter-spacing: -0.45px;\" rel=\"nofollow noopener\" target=\"_blank\">than we thought<\/a>, playing critical roles in gene regulation.<\/p>\n<p>One category of non-coding DNA is the <a href=\"https:\/\/en.wikipedia.org\/wiki\/Transposable_element\" rel=\"nofollow noopener\" target=\"_blank\">transposable element<\/a> (TE): sequences that can cut themselves free from one part of the genome and insert themselves into another.<\/p>\n<p>Related: <a href=\"https:\/\/www.sciencealert.com\/mitochondria-dump-their-rubbish-dna-and-it-could-be-costing-us-our-health\" rel=\"nofollow noopener\" target=\"_blank\">Mitochondria Dump Their Rubbish DNA, And It Could Be Costing Us Our Health<\/a><\/p>\n<p>An international team led by researchers from King&#8217;s College London (KCL) has now found that stubbornly persistent <a href=\"https:\/\/www.sciencealert.com\/hepatitis-treatment-prevents-the-progress-of-a-common-blood-cancer\" rel=\"nofollow noopener\" target=\"_blank\">blood cancers<\/a> can &#8216;wake up&#8217; TEs as part of the mechanisms that lead to  <a href=\"https:\/\/www.sciencealert.com\/cancer\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73077\" data-postid=\"179304\" rel=\"nofollow noopener\" target=\"_self\">cancer<\/a> cells going awry \u2013 and that this same activity could potentially be targeted to shut the cancers down.<a href=\"https:\/\/www.sciencealert.com\/mitochondria-dump-their-rubbish-dna-and-it-could-be-costing-us-our-health\" rel=\"nofollow noopener\" target=\"_blank\"><\/p>\n<p>More research is needed to validate these findings, which come from experiments with lab-grown cells, but unlocking a new pathway to target blood cancers could lead to treatments for cancers harboring specific mutations.<\/p>\n<p>&#8220;This discovery offers new hope for patients with hard-to-treat cancers, by using existing drugs in a completely new way, turning what was once thought to be useless DNA into a powerful target for treatment,&#8221; <a href=\"https:\/\/www.kcl.ac.uk\/news\/research-uncovers-new-treatments-for-hard-to-treat-blood-cancers\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> biologist Chi Wai Eric So, from KCL.<\/p>\n<p>The research focuses on two blood cancers, <a href=\"https:\/\/en.wikipedia.org\/wiki\/Myelodysplastic_syndrome\" rel=\"nofollow noopener\" target=\"_blank\">myelodysplastic syndrome<\/a> and <a href=\"https:\/\/www.mayoclinic.org\/diseases-conditions\/chronic-lymphocytic-leukemia\/symptoms-causes\/syc-20352428\" rel=\"nofollow noopener\" target=\"_blank\">chronic lymphocytic leukemia<\/a>. Mutations typically found in these cancers damage the genes ASXL1 and EXH2, affecting protein production and leading to uncontrolled cell growth. A cascade of instability then follows.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/ie\/wp-content\/uploads\/2025\/11\/CancerSuppression.jpg\" alt=\"DNA cancer treatment graphic\" width=\"642\" height=\"241\" class=\"wp-image-179310 size-full\"   loading=\"lazy\"\/>The researchers blocked the repair system used by cancer cells to fix DNA damage (top row), killing off the cancer. (Zeisig et al., Blood, 2025)<\/p>\n<p>What makes targeting these cancers particularly tricky is that the mutated genes no longer produce proteins that conventional cancer therapies would target.<\/p>\n<p>Using mouse models of cancer and human <a href=\"https:\/\/www.sciencealert.com\/new-breakthrough-treatment-safely-kills-cancer-cells-with-light\" rel=\"nofollow noopener\" target=\"_blank\">cancer cells<\/a>, the researchers found another knock-on effect of ASXL1 and EXH2 damage: reactivated junk DNA duplicating and spreading by inserting their sequences throughout the cancer cells&#8217; DNA.<\/p>\n<p>This unfettered behavior stresses the cancer cells. To resist that stress and continue growing, the cancer becomes reliant on <a href=\"https:\/\/en.wikipedia.org\/wiki\/Poly_(ADP-ribose)_polymerase\" rel=\"nofollow noopener\" target=\"_blank\">poly (ADP-ribose) polymerase<\/a> repair proteins \u2013 or PARPs for short. <a href=\"https:\/\/en.wikipedia.org\/wiki\/PARP_inhibitor\" rel=\"nofollow noopener\" target=\"_blank\">Drugs that suppress PARPs<\/a> were shown to be effective at killing off the two tested blood cancers. What&#8217;s more, healthy cells were mostly left alone.<\/p>\n<p>&#8220;This study sets the stage for a novel and broader approach of creating synthetic lethality for human cancers,&#8221; <a href=\"https:\/\/doi.org\/10.1182\/blood.2025028560\" rel=\"nofollow noopener\" target=\"_blank\">write<\/a> the researchers in their published paper.<\/p>\n<p>The researchers are confident their findings apply to other <a href=\"https:\/\/www.sciencealert.com\/new-report-links-alcohol-to-six-major-cancer-types\" rel=\"nofollow noopener\" target=\"_blank\">types of cancers<\/a> as well, not least because PARP blockers are already used to fight other forms of cancer, although the exact mechanisms involved differ.<br \/><a href=\"https:\/\/www.sciencealert.com\/spark-into-space-comp\" rel=\"nofollow noopener\" target=\"_blank\"><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/ie\/wp-content\/uploads\/2025\/10\/Mid-Article-Promo-Astro-642x272.jpg\" alt=\"Win a $10,000 Space Coast Adventure Holiday\" width=\"642\" height=\"272\" class=\"alignnone wp-image-177074 size-medium\"   loading=\"lazy\"\/><\/a><\/p>\n<p>And it&#8217;s another example of the hidden activity of TEs, previously dismissed as unimportant.
