{"id":373962,"date":"2026-03-31T02:24:08","date_gmt":"2026-03-31T02:24:08","guid":{"rendered":"https:\/\/www.newsbeep.com\/ie\/373962\/"},"modified":"2026-03-31T02:24:08","modified_gmt":"2026-03-31T02:24:08","slug":"new-alzheimers-treatment-strategy-reverses-cognitive-decline-in-mice-sciencealert","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/ie\/373962\/","title":{"rendered":"New Alzheimer&#8217;s Treatment Strategy Reverses Cognitive Decline in Mice : ScienceAlert"},"content":{"rendered":"<p>Researchers have developed a novel compound that could transform the way we treat  <a href=\"https:\/\/www.sciencealert.com\/go\/IaO\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73015\" data-postid=\"196191\" rel=\"nofollow noopener\" target=\"_self\">Alzheimer&#8217;s<\/a> disease, offering not just a new weapon but potentially a new strategy for battling the most common form of dementia worldwide.<\/p>\n<p>While current drugs for Alzheimer&#8217;s mostly focus on removing <a href=\"https:\/\/doi.org\/10.1038\/s41380-021-01249-0\" rel=\"nofollow noopener\" target=\"_blank\">amyloid-beta plaques<\/a> associated with the disease, the new compound takes a fundamentally different approach, instead targeting a specific enzyme to therapeutically reprogram the <a href=\"https:\/\/www.sciencealert.com\/epigenetic-changes\" rel=\"nofollow noopener\" target=\"_blank\">epigenome<\/a> of neurons \u2013 a series of molecular marks that can be added to or removed from DNA, to <a href=\"https:\/\/www.sciencealert.com\/scientists-identify-an-epigenetic-switch-that-can-slow-production-of-fat-cells\" rel=\"nofollow noopener\" target=\"_blank\">change the way genes work<\/a>.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/new-alzheimers-treatment-slows-decline-but-comes-at-a-high-cost\" rel=\"nofollow noopener\" target=\"_blank\">Monoclonal antibody<\/a> drugs such as lecanemab and donanemab, which target amyloid-beta proteins, help somewhat to slow the progression of the disease when treatment is started early, but there is still no proven way to reverse cognitive decline from Alzheimer&#8217;s in humans.<\/p>\n<p>Therapies targeting another protein called tau <a href=\"https:\/\/www.nature.com\/articles\/s41582-023-00883-2\" rel=\"nofollow noopener\" target=\"_blank\">aren&#8217;t proving very effective<\/a>, either.<\/p>\n<p>It&#8217;s led researchers to suspect we might be <a href=\"https:\/\/www.sciencealert.com\/sky-high-levels-of-alzheimers-protein-found-in-newborns\" rel=\"nofollow noopener\" target=\"_blank\">thinking about Alzheimer&#8217;s disease<\/a> <a href=\"https:\/\/www.sciencealert.com\/worlds-leading-theory-on-alzheimers-in-crisis-after-major-drug-trials-fail\" rel=\"nofollow noopener\" target=\"_blank\">all wrong<\/a> \u2013 focusing on proteins that are a sign of the disease, not the root cause of it.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/ie\/wp-content\/uploads\/2026\/03\/amyloid-beta-alzheimers-642x361.jpg\" alt=\"illustration of amyloid beta plaques in Alzheimer's disease\" width=\"642\" height=\"361\" class=\"size-medium wp-image-196290\"   loading=\"lazy\"\/>In Alzheimer&#8217;s, abnormal proteins clump together to form plaques between cells (brown) and inside cells (blue). work in a broader way, zooming out to target upstream changes in gene expression that help fuel the disease&#8217;s progression in multiple ways, not just via protein plaques.<\/p>\n<p>This hints at a new epigenetic strategy for treating Alzheimer&#8217;s disease, <a href=\"https:\/\/www.neurociencies.ub.edu\/researchers-design-a-pioneering-drug-capable-of-reversing-cognitive-decline-in-alzheimers-disease-in-animal-models\/\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> first author Aina Bellver-Sanchis, a molecular biologist at the University of Barcelona Institute of Neurosciences in Spain.<\/p>\n<p>&#8220;The compound FLAV-27 represents an innovative and promising approach to Alzheimer&#8217;s disease,&#8221; <a href=\"https:\/\/www.neurociencies.ub.edu\/researchers-design-a-pioneering-drug-capable-of-reversing-cognitive-decline-in-alzheimers-disease-in-animal-models\/\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> Bellver-Sanchis, &#8220;with the potential to modify the disease process, as it acts not only on its symptoms or a single pathological biomarker, but directly on its underlying molecular mechanisms.&#8221;<\/p>\n<p>Bellver-Sanchis and her colleagues <a href=\"https:\/\/www.neurociencies.ub.edu\/researchers-design-a-pioneering-drug-capable-of-reversing-cognitive-decline-in-alzheimers-disease-in-animal-models\/\" rel=\"nofollow noopener\" target=\"_blank\">note<\/a> that while monoclonal antibodies represent a genuine breakthrough in Alzheimer&#8217;s treatment, lecanemab and donanemab only slow cognitive decline by around 30 percent and address only part of the condition&#8217;s pathology.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/newsletter?utm_source=promo_generic_health\" rel=\"nofollow noopener\" target=\"_blank\"><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/ie\/wp-content\/uploads\/2025\/11\/Generic-Health-Promo-Final-642x273.jpg\" alt=\"Subscribe to ScienceAlert's free fact-checked newsletter\" width=\"642\" height=\"273\" class=\"alignnone wp-image-182810 size-medium\"   loading=\"lazy\"\/><\/a><\/p>\n<p>FLAV-27 is the first inhibitor of its kind to target an enzyme called <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/gene?Db=gene&amp;Cmd=ShowDetailView&amp;TermToSearch=10919\" rel=\"nofollow noopener\" target=\"_blank\">euchromatic histone-lysine N-methyltransferase 2<\/a> (EHMT2), also known as G9a.<\/p>\n<p>G9a is involved in epigenetic regulation within the brain, and can silence genes important for key tasks such as brain cell development, synaptic plasticity, and memory processing.<\/p>\n<p>FLAV-27 inhibits G9a by blocking a molecule known as <a href=\"https:\/\/doi.org\/10.1039\/d2np00086e\" rel=\"nofollow noopener\" target=\"_blank\">S-adenosylmethionine<\/a>, without which the enzyme loses its influence over genetic expression, the researchers explain.<\/p>\n<p>Inhibiting G9a seems to help calm the epigenetic dysregulation <a href=\"https:\/\/doi.org\/10.3389\/fnagi.2022.911635\" rel=\"nofollow noopener\" target=\"_blank\">seen in Alzheimer&#8217;s disease<\/a>, they report, and restores a more typical function to brain cells.<\/p>\n<p>The new compound has not yet been tested in humans \u2013 and may not be for a while \u2013 but it has shown promising results in cell experiments as well as studies on nematodes and mice.<\/p>\n<p>Beyond reducing amyloid-beta plaques and <a href=\"https:\/\/doi.org\/10.3390\/ijms232112841\" rel=\"nofollow noopener\" target=\"_blank\">tangled tau<\/a> in lab-grown mouse brain cells, FLAV-27 showed signs that it can help restore some of the damage wrought by Alzheimer&#8217;s disease in another animal model, <a href=\"https:\/\/en.wikipedia.org\/wiki\/Caenorhabditis_elegans\" rel=\"nofollow noopener\" target=\"_blank\">Caenorhabditis elegans<\/a>.<\/p>\n<p>In these nematode worms, the compound improved mobility, extended lifespan, and ramped up mitochondrial respiration, which helps fuel cells.<\/p>\n<p>In mouse models of both early- and late-onset Alzheimer&#8217;s, FLAV-27 also restored memory performance, social behavior, and the function of signalling hubs called <a href=\"https:\/\/www.sciencealert.com\/a-hidden-brain-process-may-help-explain-why-alzheimers-risk-rises-with-age\" rel=\"nofollow noopener\" target=\"_blank\">synapses<\/a> that connect brain cells.<\/p>\n<p>Related: <a href=\"https:\/\/www.neurociencies.ub.edu\/researchers-design-a-pioneering-drug-capable-of-reversing-cognitive-decline-in-alzheimers-disease-in-animal-models\/\" rel=\"nofollow noopener\" target=\"_blank\">suggest<\/a>, hints that epigenetic dysregulation could be a central mechanism linking the various pathological markers of Alzheimer&#8217;s, not just a side effect of the disease.<\/p>\n<p>The new compound shows potential, just like <a href=\"https:\/\/www.sciencealert.com\/new-alzheimers-treatment-clears-plaques-from-brains-of-mice-within-hours\" rel=\"nofollow noopener\" target=\"_blank\">other drug candidates<\/a>, but still faces a long road before human trials can begin, the team <a href=\"https:\/\/www.neurociencies.ub.edu\/researchers-design-a-pioneering-drug-capable-of-reversing-cognitive-decline-in-alzheimers-disease-in-animal-models\/\" rel=\"nofollow noopener\" target=\"_blank\">notes<\/a>, including toxicology studies in at least two animal species and other regulatory steps.<\/p>\n<p>The study was published in <a
