{"id":489803,"date":"2026-06-09T00:31:09","date_gmt":"2026-06-09T00:31:09","guid":{"rendered":"https:\/\/www.newsbeep.com\/ie\/489803\/"},"modified":"2026-06-09T00:31:09","modified_gmt":"2026-06-09T00:31:09","slug":"scientists-found-2-existing-drugs-could-reverse-alzheimers-brain-damage-in-mice-sciencealert","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/ie\/489803\/","title":{"rendered":"Scientists Found 2 Existing Drugs Could Reverse Alzheimer&#8217;s Brain Damage in Mice : ScienceAlert"},"content":{"rendered":"<p>Scientists looking to combat  <a href=\"https:\/\/www.sciencealert.com\/go\/IaO\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73015\" data-postid=\"204259\" rel=\"nofollow noopener\" target=\"_self\">Alzheimer&#8217;s<\/a> disease may have found two promising candidates.<\/p>\n<p>Better yet, they&#8217;re existing drugs that are currently used to treat  <a href=\"https:\/\/www.sciencealert.com\/cancer\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73077\" data-postid=\"204259\" rel=\"nofollow noopener\" target=\"_self\">cancer<\/a>.<\/p>\n<p>Already approved by regulators in the US \u2013 meaning potential  <a href=\"https:\/\/www.sciencealert.com\/clinical-trials\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73032\" data-postid=\"204259\" rel=\"nofollow noopener\" target=\"_self\">clinical trials<\/a> for Alzheimer&#8217;s could start sooner \u2013 the drugs are <a href=\"https:\/\/en.wikipedia.org\/wiki\/Letrozole\" rel=\"nofollow noopener\" target=\"_blank\">letrozole<\/a> (usually used to treat breast cancer) and <a href=\"https:\/\/en.wikipedia.org\/wiki\/Irinotecan\" rel=\"nofollow noopener\" target=\"_blank\">irinotecan<\/a> (usually used to treat colon and lung cancer).<\/p>\n<p>The team of US researchers started by investigating how Alzheimer&#8217;s altered <a href=\"https:\/\/en.wikipedia.org\/wiki\/Gene_expression\" rel=\"nofollow noopener\" target=\"_blank\">gene expression<\/a> in the brain.<\/p>\n<p>They then searched a medical database called the <a href=\"https:\/\/www.broadinstitute.org\/connectivity-map-cmap\" rel=\"nofollow noopener\" target=\"_blank\">Connectivity Map<\/a> for drugs that reversed these changes in <a href=\"https:\/\/www.sciencealert.com\/alcohol-changes-how-your-brains-genes-work-changing-them-back-may-fight-addiction\" rel=\"nofollow noopener\" target=\"_blank\">gene expression<\/a>, as well as cross-referenced records of patients who had taken these medicines as part of cancer treatments. The drugs appeared to have decreased their risk of developing Alzheimer&#8217;s.<\/p>\n<p>&#8220;Alzheimer&#8217;s disease comes with complex changes to the brain, which has made it tough to study and treat, but our computational tools opened up the possibility of tackling the complexity directly,&#8221; <a href=\"https:\/\/www.ucsf.edu\/news\/2025\/07\/430386\/do-these-two-cancer-drugs-have-what-it-takes-beat-alzheimers\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> computational biologist Marina Sirota, from UC San Francisco.<\/p>\n<p>&#8220;We&#8217;re excited that our computational approach led us to a potential combination therapy for Alzheimer&#8217;s based on existing FDA-approved medications.&#8221;<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/ie\/wp-content\/uploads\/2026\/06\/Screenshot-2025-07-24-at-4.31.54\u202fpm-642x494.png\" alt=\"Certain Cancer Drugs May Reverse Brain Changes That Occur During Alzheimer's\" width=\"642\" height=\"494\" class=\"wp-image-169045 size-medium\"   loading=\"lazy\"\/>The application of letrozole and irinotecan reduced the level of tau proteins (light green) in mouse brains. (Li et al., Cell, 2025)<\/p>\n<p>Having picked out letrozole and irinotecan as the best candidates, the researchers tested them in mouse models of Alzheimer&#8217;s. When used in tandem, the drugs were shown to reverse some of the brain changes <a href=\"https:\/\/www.sciencealert.com\/alzheimers-disease-harms-the-brain-in-2-distinct-phases-study-reveals\" rel=\"nofollow noopener\" target=\"_blank\">brought on by the disease<\/a>.<\/p>\n<p>The harmful clumps of <a href=\"https:\/\/en.wikipedia.org\/wiki\/Tau_protein\" rel=\"nofollow noopener\" target=\"_blank\">tau protein<\/a> that build up in brains affected by Alzheimer&#8217;s were reduced significantly, and the mice showed improvements in learning and memory tasks \u2013 two brain capabilities often impaired by Alzheimer&#8217;s.<\/p>\n<p>By combining the two drugs, the researchers were able to target different types of brain cells affected by the disease. Letrozole seemed to counter Alzheimer&#8217;s in neurons, while irinotecan worked in <a href=\"https:\/\/en.wikipedia.org\/wiki\/Glia\" rel=\"nofollow noopener\" target=\"_blank\">glia<\/a>.<\/p>\n<p>&#8220;Alzheimer&#8217;s is likely the result of numerous alterations in many genes and proteins that, together, disrupt brain health,&#8221; <a href=\"https:\/\/www.ucsf.edu\/news\/2025\/07\/430386\/do-these-two-cancer-drugs-have-what-it-takes-beat-alzheimers\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> neuroscientist Yadong Huang, from UC San Francisco and Gladstone Institutes.<\/p>\n<p>&#8220;This makes it very challenging for drug development \u2013 which traditionally produces one drug for a single gene or protein that drives disease.&#8221;<\/p>\n<p>It&#8217;s a promising start, but there&#8217;s more work to be done: Obviously the drugs have only been directly tested in mice so far, and these medications also come with <a href=\"https:\/\/www.nhs.uk\/medicines\/letrozole\/side-effects-of-letrozole\/\" rel=\"nofollow noopener\" target=\"_blank\">side effects<\/a>. These need to be considered alongside the benefits if the drugs are to be <a href=\"https:\/\/www.sciencealert.com\/this-common-blood-pressure-drug-extends-lifespan-slows-aging-in-animals\" rel=\"nofollow noopener\" target=\"_blank\">repurposed for a different disease<\/a> than what they were originally approved for.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/newsletter?utm_source=promo_stargazing\" rel=\"nofollow noopener\" target=\"_blank\"><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/ie\/wp-content\/uploads\/2026\/06\/FOMO-1200x628-_Dont-miss-the-next-breakthrough_-cosmic-colourful-642x336.jpg\" alt=\"Subscribe to ScienceAlert's free fact-checked newsletter\" width=\"642\" height=\"336\" class=\"alignnone wp-image-201722 size-medium\"   loading=\"lazy\"\/><\/a><\/p>\n<p>One of the next steps should be clinical trials for people with <a href=\"https:\/\/www.sciencealert.com\/theres-a-critical-thing-we-can-all-do-to-hold-alzheimers-symptoms-at-bay\" rel=\"nofollow noopener\" target=\"_blank\">Alzheimer&#8217;s disease<\/a>. According to the researchers, this approach could lead to more personalized and effective treatments, based on how gene expression has been altered in each case.<\/p>\n<p>It&#8217;s estimated that <a href=\"https:\/\/www.alzint.org\/about\/dementia-facts-figures\/dementia-statistics\/\" rel=\"nofollow noopener\" target=\"_blank\">more than 55 million people<\/a> have Alzheimer&#8217;s, and as the world&#8217;s population ages, that&#8217;s expected to more than double in the next 25 years.
