{"id":588314,"date":"2026-08-09T21:56:10","date_gmt":"2026-08-09T21:56:10","guid":{"rendered":"https:\/\/www.newsbeep.com\/ie\/588314\/"},"modified":"2026-08-09T21:56:10","modified_gmt":"2026-08-09T21:56:10","slug":"experimental-alzheimers-treatment-restores-two-hours-of-sleep-when-tested-in-mice-sciencealert","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/ie\/588314\/","title":{"rendered":"Experimental Alzheimer&#8217;s Treatment Restores Two Hours of Sleep When Tested in Mice : ScienceAlert"},"content":{"rendered":"<p><a href=\"https:\/\/www.sciencealert.com\/alzheimer-s-disease\" rel=\"nofollow noopener\" target=\"_blank\">Alzheimer&#8217;s disease<\/a> is such a challenging condition to study because it presents researchers with a complex mesh of risk factors, causes, and symptoms that need untangling \u2013 and sleep is a prime example.<\/p>\n<p>We know that poor sleep can raise <a href=\"https:\/\/www.sciencealert.com\/one-stage-of-sleep-seems-critical-for-reducing-the-risk-of-dementia\" rel=\"nofollow noopener\" target=\"_blank\">the risk of Alzheimer&#8217;s<\/a>, and that the disease itself interferes with normal sleep processes, but understanding the various contributors and consequences at play is difficult.<\/p>\n<p>Now, we may have a bit more clarity on the relationship between  <a href=\"https:\/\/www.sciencealert.com\/go\/IaO\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73015\" data-postid=\"212645\" rel=\"nofollow noopener\" target=\"_self\">Alzheimer&#8217;s<\/a> and sleep, thanks to a new study published in <a href=\"https:\/\/doi.org\/10.1002\/alz.71579\" rel=\"nofollow noopener\" target=\"_blank\">Alzheimer&#8217;s &amp; Dementia<\/a>, led by researchers from the University of Kentucky.<\/p>\n<p>The researchers identified brain immune cells <a href=\"https:\/\/www.sciencealert.com\/switch-turns-brains-defenses-into-protectors-against-alzheimers\" rel=\"nofollow noopener\" target=\"_blank\">called microglia<\/a> as being potential candidates for stealing sleep in people with Alzheimer&#8217;s \u2013 effectively putting the brain on high alert as the disease takes hold, in a way that interferes with natural rest.<\/p>\n<p>It shifts the focus away from the toxic clumps of amyloid-beta protein that build up in the brain alongside Alzheimer&#8217;s disease progression.<\/p>\n<p>As far as disrupted sleep goes, these plaques might be the trigger, rather than what&#8217;s doing the actual damage.<\/p>\n<p><img decoding=\"async\" width=\"642\" height=\"156\" src=\"https:\/\/www.newsbeep.com\/ie\/wp-content\/uploads\/2026\/08\/MouseScans.jpg\" alt=\"Mouse scans\" class=\"wp-image-212648\"   loading=\"lazy\"\/>The researchers looked at how amyloid-beta plaques (purple) affected sleep quality through brainwave frequencies. (Constantino et al., Alzheimer&#8217;s &amp; Dementia, 2026)<\/p>\n<p>&#8220;Basically, we showed that it is not the plaques themselves, or solely dysfunctional neurons, that cause sleep loss but actually microglia,&#8221; <a href=\"https:\/\/uknow.uky.edu\/research\/two-hours-sleep-restored-uk-researchers-make-alzheimer-s-breakthrough\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> physiologist Shannon Macauley from the University of Kentucky.<\/p>\n<p>&#8220;Microglia are immune cells that, when they respond to plaques, kick off this elaborate cascade of inflammation, as if the microglia are partying all night, and keeping the brain awake.&#8221;<\/p>\n<p>The researchers compared two groups of mice for the study: mice genetically engineered to develop amyloid-beta plaques <a href=\"https:\/\/www.sciencealert.com\/common-sleeping-pill-may-reduce-buildup-of-alzheimers-proteins-study-reveals\" rel=\"nofollow noopener\" target=\"_blank\">similar to those<\/a> found in Alzheimer&#8217;s patients, and a healthy control group.<\/p>\n<p><img decoding=\"async\" width=\"721\" height=\"387\" src=\"https:\/\/www.newsbeep.com\/ie\/wp-content\/uploads\/2026\/08\/NeuronDetails.jpg\" alt=\"Neuron image analysis\" class=\"wp-image-212650\"   loading=\"lazy\"\/>Brain tissue scans, showing the effects of immune cells being turned off (right) on microglia and amyloid plaques. (Nicholas Constantino)<\/p>\n<p>Sleep cycles, brain activity, and amyloid-beta plaque build-up were monitored in the animals, specifically at six months old when the disease was just getting started, and at 18 months when it was well developed.<\/p>\n<p>The data showed that the appearance of plaques cost the mice 1.5-2 hours of <a href=\"https:\/\/www.sciencealert.com\/sticking-to-the-same-bedtime-each-night-could-help-lower-heart-health-risk\" rel=\"nofollow noopener\" target=\"_blank\">sleep per night<\/a>, right away. This was consistent between six and 18 months, even though substantially more plaque accumulated over that time.<\/p>\n<p>&#8220;I expected that as plaque burden became more severe, sleep disruption would also worsen,&#8221; <a href=\"https:\/\/uknow.uky.edu\/research\/two-hours-sleep-restored-uk-researchers-make-alzheimer-s-breakthrough\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> neuroscientist Nicholas Constantino from the University of Kentucky.<\/p>\n<p>&#8220;The disruptions in sleep and cortical <a href=\"https:\/\/en.wikipedia.org\/wiki\/Electroencephalography\" rel=\"nofollow noopener\" target=\"_blank\">EEG activity<\/a> that occur at six months, when plaques first emerge, did not worsen by 18 months, despite more than double the amount of plaque burden.&#8221;<\/p>\n<p>In a subsequent experiment based on a similar premise with a different group of mice, the researchers introduced dosages of microglia-blocking drugs.<\/p>\n<p>This returned the headline finding: disabling the microglia reaction gave the Alzheimer&#8217;s mice an extra two hours of sleep per night, extending their <a href=\"https:\/\/www.sciencealert.com\/sleeping-less-than-6-hours-a-night-linked-to-higher-dementia-risk-scientists-show\" rel=\"nofollow noopener\" target=\"_blank\">restorative sleep periods<\/a>.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/newsletter?int_promo=stargazing\" rel=\"nofollow noopener\" target=\"_blank\"><img decoding=\"async\" width=\"642\" height=\"336\" src=\"https:\/\/www.newsbeep.com\/ie\/wp-content\/uploads\/2026\/06\/FOMO-1200x628-_Dont-miss-the-next-breakthrough_-cosmic-colourful-642x336.jpg\" alt=\"Subscribe to ScienceAlert's free fact-checked newsletter\" class=\"wp-image-201722\"   loading=\"lazy\"\/><\/a><\/p>\n<p>&#8220;That restorative sleep is super important for physical repair, learning and memory and washing out the toxins of the day,&#8221; <a href=\"https:\/\/uknow.uky.edu\/research\/two-hours-sleep-restored-uk-researchers-make-alzheimer-s-breakthrough\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> Macauley.<\/p>\n<p>&#8220;When Alzheimer&#8217;s patients lose this stage, they lose their brain&#8217;s primary cleaning cycle, creating a feed-forward loop that may drive further damage.&#8221;<\/p>\n<p>The drug dosage had no effect on the amyloid-beta plaques, which were unchanged. It&#8217;s previously <a href=\"https:\/\/doi.org\/10.1001\/jamaneurol.2021.2876\" rel=\"nofollow noopener\" target=\"_blank\">been suggested<\/a> that the death of neurons in the brain or the accumulation of amyloid-beta proteins is what&#8217;s causing poor sleep in Alzheimer&#8217;s patients.<\/p>\n<p>By tackling the sleep problem through microglia, we could make some progress on slowing down or <a href=\"https:\/\/www.sciencealert.com\/new-drug-stalls-alzheimers-development-in-breakthrough-trial\" rel=\"nofollow noopener\" target=\"_blank\">limiting Alzheimer&#8217;s<\/a>.<\/p>\n<p>Clearly there&#8217;s a lot of work to do \u2013 this has only been tested in mice so far, and wiping out an army of brain immune cells isn&#8217;t really an option.
