Semaglutide\u2014better known as Ozempic and Wegovy\u2014seems to be everywhere. The blockbuster weight loss aid mimics a natural hormone that tells the brain \u201cyou\u2019re full,\u201d making it easier to shed pounds without the constant hunger and cravings that make traditional dieting miserable.<\/p>\n<p class=\"css-1xcfe9l\">The drug may also have longer effects. <a href=\"https:\/\/www.nature.com\/articles\/s41586-026-10940-7\" rel=\"nofollow noopener\" target=\"_blank\">A new study<\/a> in mice suggests that starting semaglutide in old age extends lifespan roughly 12 percent. The mice regained their curiosity and memory and improved on myriad age-related hallmarks. Chronic inflammation cooled. Senescent \u201czombie cells\u201d dwindled. Their genomes and proteins became more stable, and the hippocampus\u2014a brain region crucial for learning and memory\u2014sprouted new neurons.<\/p>\n<p class=\"css-1xcfe9l\">The findings are reminiscent of calorie restriction, a way to boost longevity and stave off age-related health problems, at least in lab animals. But sticking to a diet for years, let alone decades, is tough. Scientists have long searched for a drug that could deliver some of the benefits without hunger pangs. Semaglutide seems to fit the bill, with surprising perks beyond dieting.<\/p>\n<p class=\"css-1xcfe9l\">\u201cIf these results from mice hold true in people, this is a promising hint that people taking GLP-1R agonists [GLP-1 drugs] may have an age-slowing benefit as well as benefits to reducing diabetes and obesity,\u201d <a href=\"https:\/\/www.sciencemediacentre.org\/expert-reaction-to-glp-1r-activating-drug-treatment-slows-ageing-and-extends-lifespan-in-female-mice\/\" rel=\"nofollow noopener\" target=\"_blank\">said<\/a> Tara Spires-Jones at the University of Edinburgh, who was not involved in the study.<\/p>\n<p class=\"css-1xcfe9l\">But don\u2019t go ordering GLP-1 drugs online just yet. The study used inbred female mice, whose physiology differs from that of women aging through or beyond menopause. Rapid weight loss in people taking these drugs can also reduce lean muscle, potentially increasing fragility in older people. And while GLP-1 drugs are already being tested in the battle against neurodegenerative disorders, whether they sharpen the aging brain remains an open question.<\/p>\n<p class=\"css-1xcfe9l\">Still, longevity researchers are cautiously optimistic.<\/p>\n<p class=\"css-1xcfe9l\">\u201cThe findings reframe a key question that has often been asked back to front,\u201d <a href=\"https:\/\/www.nature.com\/articles\/d41586-026-02468-7\" rel=\"nofollow noopener\" target=\"_blank\">wrote<\/a> Maria Fernandez and Rafael de Cabo at the National Institute of Aging, who were not involved in the study. \u201cRather than considering the broad health benefits of GLP-1 drugs as something to be explained one disease at a time, these results suggest that the positive effects have a common cause: slowed aging.\u201d<\/p>\n<p>Fountain of Youth<\/p>\n<p class=\"css-1xcfe9l\">One way to live healthier and longer is seemingly mundane: adopt a healthy lifestyle.<\/p>\n<p class=\"css-1xcfe9l\">Diet and exercise have repeatedly been linked to better health in our twilight years. As we age, our bodies slowly break down. Damage and mutations accumulate in DNA. Telomeres, the protective caps at the ends of chromosomes, grow shorter, contributing to genomic instability. The molecular switches that turn genes on or off go haywire, and our cells become less able to make working proteins and clean up damaged ones.<\/p>\n<p class=\"css-1xcfe9l\">There\u2019s more. Mitochondria, the cell\u2019s power plants, struggle to produce energy and leak toxic molecules. Damaged cells stop dividing, lose their function, but stubbornly refuse to die. Instead, they leak a toxic chemical soup that damages tissues. Chronic inflammation flares, and stem cells run out of steam, making it harder for the body to regenerate or repair itself.<\/p>\n<p class=\"css-1xcfe9l\">Together called the <a href=\"https:\/\/www.sciencedirect.com\/science\/article\/pii\/S0092867422013770\" rel=\"nofollow noopener\" target=\"_blank\">hallmarks of aging<\/a>, this laundry list has long challenged scientists looking for a silver bullet against the march of time. They have discovered some exotic options. Transferring components of young mice\u2019s blood to older recipients has rejuvenated faltering <a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC3677132\/\" rel=\"nofollow noopener\" target=\"_blank\">hearts<\/a>, <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/29040401\/\" rel=\"nofollow noopener\" target=\"_blank\">kidneys<\/a>, and <a href=\"https:\/\/www.nature.com\/articles\/nm.3569\" rel=\"nofollow noopener\" target=\"_blank\">brains<\/a>. And clearing senescent \u201czombie\u201d cells with <a href=\"https:\/\/singularityhub.com\/2023\/11\/07\/zombie-cells-have-a-weakness-a-new-anti-aging-therapy-exploits-it\/\" rel=\"nofollow noopener\" target=\"_blank\">drug cocktails<\/a> or genetic engineering has attracted <a href=\"https:\/\/www.newyorker.com\/magazine\/2017\/04\/03\/silicon-valleys-quest-to-live-forever\" rel=\"nofollow noopener\" target=\"_blank\">billions of dollars<\/a> in investment.<\/p>\n<p class=\"css-1xcfe9l\">But perhaps the most studied, and most robust, intervention is <a href=\"https:\/\/www.nature.com\/articles\/s43587-026-01091-5\" rel=\"nofollow noopener\" target=\"_blank\">caloric restriction<\/a>. In flies, rodents, and other lab animals, slashing energy intake without causing malnutrition has repeatedly extended lifespan and delayed multiple age-related diseases. Restriction triggers broad changes, including improved metabolism and insulin sensitivity. It also helps prevent damage to cells. In humans, <a href=\"https:\/\/singularityhub.com\/2023\/02\/21\/cutting-out-just-a-muffin-a-day-can-slow-aging-study-finds\/\" rel=\"nofollow noopener\" target=\"_blank\">moderately reducing calories<\/a> seems to improve heart health and slow the speed of biological aging.<\/p>\n<p class=\"css-1xcfe9l\">Sticking to a diet for years on end, however, is hardly sustainable.<\/p>\n<p>Cheat Code<\/p>\n<p class=\"css-1xcfe9l\">GLP-1 drugs may make it easier.<\/p>\n<p class=\"css-1xcfe9l\">Originally designed to control blood sugar and appetite, the drugs have also shown promise for reducing rates of <a href=\"https:\/\/www.fda.gov\/news-events\/press-announcements\/fda-approves-first-treatment-reduce-risk-serious-heart-problems-specifically-adults-obesity-or\" rel=\"nofollow noopener\" target=\"_blank\">cardiovascular<\/a>, <a href=\"https:\/\/www.nejm.org\/doi\/abs\/10.1056\/NEJMoa2403347\" rel=\"nofollow noopener\" target=\"_blank\">kidney<\/a>, <a href=\"https:\/\/www.fda.gov\/drugs\/news-events-human-drugs\/fda-approves-treatment-serious-liver-disease-known-mash\" rel=\"nofollow noopener\" target=\"_blank\">liver<\/a>, and <a href=\"https:\/\/www.sciencedirect.com\/science\/article\/pii\/S0091302225000433\" rel=\"nofollow noopener\" target=\"_blank\">neurodegenerative diseases<\/a>, at least in people with obesity or Type 2 diabetes. <a href=\"https:\/\/www.nature.com\/articles\/s41514-026-00383-9\" rel=\"nofollow noopener\" target=\"_blank\">Clinical trials<\/a> are now exploring their effects in people with metabolic liver disease, which becomes more common and consequential with age.<\/p>\n<p class=\"css-1xcfe9l\">Not all these effects can be explained <a href=\"https:\/\/www.nejm.org\/doi\/abs\/10.1056\/NEJMra2500106\" rel=\"nofollow noopener\" target=\"_blank\">solely by weight loss<\/a>, raising a bigger question: How can a single class of drugs influence so many seemingly unrelated conditions that <a href=\"https:\/\/www.nature.com\/articles\/s41587-025-02932-1\" rel=\"nofollow noopener\" target=\"_blank\">often crop up with age<\/a>?<\/p>\n<p class=\"css-1xcfe9l\">\u201cIf GLP-1 medications slow down the aging process itself, a wide range of clinical benefits is<\/p>\n<p class=\"css-1xcfe9l\">exactly what would be expected, because aging is the root of most chronic diseases,\u201d wrote Fernandez and de Cabo.<\/p>\n<p class=\"css-1xcfe9l\">To test that theory, the team gave daily semaglutide injections to 20-month-old female mice\u2014roughly comparable to women in their <a href=\"https:\/\/pmc.ncbi.nlm.nih.gov\/articles\/PMC5367550\/\" rel=\"nofollow noopener\" target=\"_blank\">early-to-mid 60s<\/a>\u2014for as long as they lived.<\/p>\n<p class=\"css-1xcfe9l\">Compared to a group of mice given saline, the treated mice lived an average of 834 days, versus 724 days for a control group. Both groups could feast on standard chow to their hearts\u2019 content. After just three months of treatment, the mice taking semaglutide were more lively and curious than their peers. They readily explored new environments, balanced better on a skinny rotating rod, ran faster on a tiny treadmill, and solved mazes more quickly.<\/p>\n<p class=\"css-1xcfe9l\">Under the hood, semaglutide blunted the hallmarks of aging across the board. Stem cells in the bone marrow and hippocampus sprouted, suggesting renewed regenerative capability. DNA damage and protein and energy dysfunction declined. Zombie cells partially disappeared.<\/p>\n<p class=\"css-1xcfe9l\">The results weren\u2019t simply a consequence of eating less. In another three-month experiment, the team compared the drug with a calorie-restricted diet that cut energy intake by 24 percent\u2014the same reduction seen in the semaglutide-treated mice.<\/p>\n<p class=\"css-1xcfe9l\">The drug seemed to have a leg up. Compared with caloric restriction, it produced more improvement in cognition and blood sugar control, without the metabolic adaptations normally driven by hunger, such as lowered energy expenditure. The mice also showed fewer signs of hunger. They ate on a normal schedule rather than prowling for food before feeding time and gobbling rations once available.<\/p>\n<p class=\"css-1xcfe9l\">Genetic sequencing of the liver found semaglutide triggered similar molecular signaling pathways as caloric restriction, like for example, those that sense nutrient availability, cell stress, and <a href=\"https:\/\/onlinelibrary.wiley.com\/doi\/abs\/10.1002\/jbt.23812\" rel=\"nofollow noopener\" target=\"_blank\">proteins involved in longevity<\/a>. Both interventions tamped down inflammation and boosted genes involved in handling fats, but they didn\u2019t follow the <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/30442801\/\" rel=\"nofollow noopener\" target=\"_blank\">exact same biological playbook<\/a>.<\/p>\n<p class=\"css-1xcfe9l\">The findings raise the \u201cintriguing question of whether GLP-1 drugs target an alternative biological route into aging that has its own side effects and therapeutic ceiling,\u201d wrote Fernandez and de Cabo. Exactly how that route works remains unclear, but the team is eager to find out.<\/p>\n<p class=\"css-1xcfe9l\">The findings are promising, but the study also has major limitations. It didn\u2019t directly compare dieting and semaglutide for lifespan extension. And because gender affects the aging process, the team will need to see if the results hold in males. Then there\u2019s the potential loss of lean muscle, a serious concern for people who are already frail or saddled with age-related health conditions.<\/p>\n<p class=\"css-1xcfe9l\">Even so, \u201csemaglutide is probably the best caloric-restriction mimetic I have seen,\u201d Tim Rhoads at the University of Wisconsin\u2013Madison , who was not involved in the study, told <a href=\"https:\/\/cen.acs.org\/biological-chemistry\/semaglutide-mouse-longevity\/104\/web\/2026\/09\" rel=\"nofollow noopener\" target=\"_blank\">Chemical and Engineering News<\/a>.<\/p>\n<p class=\"css-1xcfe9l\">Untangling semaglutide\u2019s bonus effects could ultimately reveal new ways to slow\u2014or even rewind\u2014aging\u2019s ticking clock.<\/p>\n","protected":false},"excerpt":{"rendered":"It\u2019s a hot GLP-1 drug summer.
