{"id":136398,"date":"2025-11-16T14:03:14","date_gmt":"2025-11-16T14:03:14","guid":{"rendered":"https:\/\/www.newsbeep.com\/il\/136398\/"},"modified":"2025-11-16T14:03:14","modified_gmt":"2025-11-16T14:03:14","slug":"a-crucial-genetic-mutation-behind-crohns-disease-has-finally-been-revealed","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/il\/136398\/","title":{"rendered":"A Crucial Genetic Mutation Behind Crohn&#8217;s Disease Has Finally Been Revealed"},"content":{"rendered":"<p class=\"mb-4 text-lg md:leading-8 break-words\">Mutations in a gene associated with <a href=\"https:\/\/www.sciencealert.com\/blood-tests-can-reveal-crohns-disease-8-years-before-diagnosis\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:Crohn&#039;s disease;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">Crohn&#8217;s disease<\/a> have been found to rob critical immune cells of their ability to switch modes, causing them to overreact and trigger inflammation.<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">Variations in the NOD2 gene have been linked to Crohn&#8217;s in <a href=\"https:\/\/doi.org\/10.1038\/s41598-021-84938-8\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:previous studies;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">previous studies<\/a>, yet their exact role in the disease&#8217;s pathology has long been a mystery.<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">Researchers from the University of California, San Diego used <a href=\"https:\/\/en.wikipedia.org\/wiki\/Machine_learning\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:machine learning;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">machine learning<\/a> techniques to identify patterns in gene activity of immune cells in the gut.<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">Experiments on lab-grown cells and samples from both healthy guts and digestive tracts with a form of Crohn&#8217;s called inflammatory bowel disease (IBD) revealed that the mutations interfere with typical protective mechanisms that allow NOD2 proteins to <a href=\"https:\/\/www.sciencealert.com\/killer-t-cells-gone-rogue-may-be-a-main-culprit-of-inflammatory-bowel-disease\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:guard against IBD;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">guard against IBD<\/a>.<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">Related: <a href=\"https:\/\/www.sciencealert.com\/these-dna-damaging-molecules-may-be-the-link-between-colon-cancer-and-ibd\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:These DNA-Damaging Molecules May Be The Link Between Colon Cancer And IBD;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">These DNA-Damaging Molecules May Be The Link Between Colon Cancer And IBD<\/a><\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">Tracking the behavior of immune cells called <a href=\"https:\/\/en.wikipedia.org\/wiki\/Macrophage\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:macrophages;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">macrophages<\/a> through the expression of their genes, the researchers discerned which cells <a href=\"https:\/\/www.sciencealert.com\/researchers-trick-a-faulty-immune-system-into-repairing-the-gut-of-mice\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:helped the gut stay healthy;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">helped the gut stay healthy<\/a> and which became inflammatory and caused damage.<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">&#8220;The gut is a battlefield, and macrophages are the peacekeepers,&#8221; <a href=\"https:\/\/today.ucsd.edu\/story\/rebalancing-the-gut-how-ai-solved-a-25-year-crohns-disease-mystery\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:says;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">says<\/a> UC San Diego Gajanan Katkar. &#8220;For the first time, AI has allowed us to clearly define and track the players on two opposing teams.&#8221;<\/p>\n<p><img alt=\"Macrophage diagram\" loading=\"lazy\" width=\"642\" height=\"636\" decoding=\"async\" data-nimg=\"1\" class=\"rounded-lg\" style=\"color:transparent\" src=\"https:\/\/www.newsbeep.com\/il\/wp-content\/uploads\/2025\/11\/f1ee76dbf9157b14be1f250fb1500b16.jpeg\"\/><\/p>\n<p>The researchers describe macrophages in the gut as being on opposing teams, increasing or reducing inflammation. (Katkar et al., J. Clin. Invest., 2025)<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">Macrophages in the gut can switch between two states, one for attacking infection (inflammatory) and one for repairing damage (non-inflammatory). It&#8217;s been well established that keeping these two states balanced is important for a <a href=\"https:\/\/www.sciencealert.com\/lifting-weights-could-change-your-gut-microbiome-within-weeks\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:healthy gut;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">healthy gut<\/a>.<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">The research team put together a genetic signature involving 53 genes, which controlled the macrophage state in IBD.<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">One gene found to promote the non-inflammatory macrophage mode produces a protein called girdin. It turns out that the girdin and NOD2 proteins work together to ensure macrophages remain alert to threats, but not overly reactive. Without them, macrophages in <a href=\"https:\/\/www.yahoo.com\/news\/biomaterial-could-finally-repair-damaged-233022279.html\" data-ylk=\"slk:repair mode;elm:context_link;itc:0;sec:content-canvas;outcm:mb_qualified_link;_E:mb_qualified_link;ct:story;\" class=\"link  yahoo-link\" rel=\"nofollow noopener\" target=\"_blank\">repair mode<\/a> are less efficient at cleaning up targets, while those in attack mode are overly inflammatory.<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">&#8220;NOD2 functions as the body&#8217;s infection surveillance system,&#8221; <a href=\"https:\/\/today.ucsd.edu\/story\/rebalancing-the-gut-how-ai-solved-a-25-year-crohns-disease-mystery\" rel=\"nofollow noopener\" target=\"_blank\" data-ylk=\"slk:says;elm:context_link;itc:0;sec:content-canvas\" class=\"link \">says<\/a> cell biologist Pradipta Ghosh, also of UC San Diego. &#8220;When bound to girdin, it detects invading pathogens and maintains gut immune balance by swiftly neutralizing them.&#8221;<\/p>\n<p class=\"mb-4 text-lg md:leading-8 break-words\">&#8220;Without this partnership, the NOD2 surveillance system collapses.&#8221;<\/p>\n<p><img alt=\"Macrophage activity in the gut\" loading=\"lazy\" width=\"642\" height=\"330\" decoding=\"async\" data-nimg=\"1\" class=\"rounded-lg\" style=\"color:transparent\" src=\"https:\/\/www.newsbeep.com\/il\/wp-content\/uploads\/2025\/11\/00fa51176ba4bbeb7954853b26c5bde9.jpeg\"\/><\/p>\n<p>Macrophages expressing girdin (left) can neutralize pathogens by forming phagolysosomes (PL).
