{"id":395351,"date":"2026-04-16T14:45:09","date_gmt":"2026-04-16T14:45:09","guid":{"rendered":"https:\/\/www.newsbeep.com\/il\/395351\/"},"modified":"2026-04-16T14:45:09","modified_gmt":"2026-04-16T14:45:09","slug":"nearly-400-compounds-affect-behaviors-tied-to-autism-linked-genes-in-zebrafish","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/il\/395351\/","title":{"rendered":"Nearly 400 compounds affect behaviors tied to autism-linked genes in zebrafish"},"content":{"rendered":"<p>Hundreds of compounds counteract specific behavioral effects of autism-linked gene variants in zebrafish, according to a new <a href=\"https:\/\/doi.org\/10.1073\/pnas.2518846123\" target=\"_blank\" rel=\"nofollow noopener\">study<\/a> that, its authors say, could guide precision gene-based medicine for some forms of autism.<\/p>\n<p>Many new drugs under investigation for the condition prove ineffective in clinical trials\u2014in part because of its genetic and clinical <a href=\"https:\/\/www.thetransmitter.org\/spectrum\/weaponized-heterogeneity-only-harms-the-most-vulnerable-autistic-people\/\" rel=\"nofollow noopener\" target=\"_blank\">heterogeneity<\/a>. \u201cA medication may be used to treat many different kinds of patients, when its use should be based on an individual\u2019s specific genes or behaviors,\u201d says study investigator <a href=\"https:\/\/medicine.yale.edu\/profile\/ellen-hoffman\/\" target=\"_blank\" rel=\"nofollow noopener\">Ellen Hoffman<\/a>, associate professor of neuroscience at Yale University\u2019s Child Study Center.\u00a0<\/p>\n<p>To address this heterogeneity, Hoffman and her colleagues developed a large-scale platform to test drugs on zebrafish with autism-linked variants and monitor their behavioral effects in an automated way. <a href=\"https:\/\/www.thetransmitter.org\/spectrum\/zebrafish-social-behavior-swims-into-mainstream\/\" rel=\"nofollow noopener\" target=\"_blank\">Zebrafish are commonly used in neuroscience<\/a> to study autism and other conditions, in part because their young are transparent, making it possible to image the whole brain in real time.<\/p>\n<p>In the new study, Hoffman and her colleagues screened 520 U.S. Food and Drug Administration-approved drugs, including antibiotics and immunosuppressants, on larval zebrafish with CRISPR-Cas-induced variants in nine genes significantly associated with autism. They developed <a href=\"http:\/\/149.165.175.233\/\" target=\"_blank\" rel=\"nofollow\">an open-source, searchable database<\/a> of the effects of these drugs on behaviors such as responses to light and darkness.<\/p>\n<p>\u201cRepurposing existing drugs really helps minimize the burden of clinical trials,\u201d notes <a href=\"https:\/\/www.umassmed.edu\/thymelab\/people\/dr.-summer-thyme\" target=\"_blank\" rel=\"nofollow noopener\">Summer Thyme<\/a>, associate professor of biochemistry and molecular biotechnology at the University of Massachusetts Chan Medical School, who was not involved in this research.<\/p>\n<p>Of the genes investigated, <a href=\"https:\/\/gene.sfari.org\/database\/human-gene\/scn2a\" target=\"_blank\" rel=\"nofollow noopener\">SCN2A<\/a> and <a href=\"https:\/\/gene.sfari.org\/database\/human-gene\/DYRK1A\" target=\"_blank\" rel=\"nofollow noopener\">DYRK1A<\/a> variants displayed the most robust behavioral effects in the fish. SCN2A variants led to nighttime hyperactivity and light hypersensitivity to lights turning on, whereas DYRK1A variants resulted in reduced sensitivity to lights turning off. Both variants also led to less activity during the day compared with wildtype fish.<\/p>\n<p><p class=\"first-letter:text-2xl first-letter:float-left first-letter:text-red first-letter:pr-2 pb-0\">\n    O<\/p>\n<p>f the 520 drugs tested, 376 displayed significant effects on at least two behaviors in fish with SCN2A or DYRK1A variants. The strongest of these compounds included estropipate, which targets an estrogen receptor and affected SCN2A variant behaviors; paclitaxel, a microtubule inhibitor that suppressed DYRK1A variant behaviors; and levocarnitine, a mitochondrial modulator and lipid metabolism regulator that targeted both kinds of variants.<\/p>\n<p>The scientists detailed their findings last month in the Proceedings of the National Academy of Sciences.\u00a0<\/p>\n<p>Previously, \u201clevocarnitine was tested in small trials for efficacy in people with autism but no genetic diagnosis and showed promise,\u201d says <a href=\"https:\/\/dallmanlab.weebly.com\/\" target=\"_blank\" rel=\"nofollow noopener\">Julia Dallman<\/a>, associate professor of biology at the University of Miami, who was not involved in this study.\u00a0<\/p>\n<p>Levocarnitine also helps treat signaling problems in human pluripotent stem cell-derived glutamatergic neurons carrying variants in SCN2A or DYRK1A, the new study showed. \u201cThese findings provide evidence for the conservation of drug rescue effects in zebrafish and human cells,\u201d Hoffman says.<\/p>\n<p>Estropipate, paclitaxel and levocarnitine did not completely reverse the behavioral effects, Thyme cautions, which suggests these variants likely affect multiple pathways\u2014something that calls for further investigation.\u00a0<\/p>\n<p class=\"font-light italic text-2sm p-0 lg:text-3md\">\n            Repurposing existing drugs really helps minimize the burden of clinical trials.         <\/p>\n<p> \u2014 <\/p>\n<p>                 \u2014<br \/>\n                Summer Thyme <\/p>\n<p>Beyond SCN2A and DYRK1A, the effort identified a notable pattern across all nine genes studied, which\u2014a past <a href=\"https:\/\/doi.org\/10.1016\/j.celrep.2023.112243\" target=\"_blank\" rel=\"nofollow noopener\">investigation<\/a> revealed\u2014cluster into three groups with distinct behavioral profiles. Psychotropic medications, such as antidepressants and antipsychotics, that benefited one particular behavioral profile, Hoffman and her colleagues found, might worsen another. For instance, mTOR antagonists reduced behaviors in two of the groups but exacerbated the behavior profile in the third. Meanwhile, dopaminergic agonists seemed to intensify the behavioral patterns of those first two groups and eased them in the third. The result underscores the need to tailor potential autism therapy regimens to a person\u2019s genetic profile, the investigators say.\u00a0<\/p>\n<p>\u201cWith further validation, therapeutic pathways identified as being effective in subtypes of autism could become a part of standard clinical care,\u201d Dallman says.<\/p>\n<p>All in all, the findings contribute to growing evidence that estrogens, microtubules, mitochondria and lipid metabolism may play central roles in some forms of autism, Hoffman says. \u201cOur results lay the groundwork for investigating the extent to which these pathways might represent targets for individuals carrying mutations in these genes.\u201d\u00a0\u00a0<\/p>\n<p>Hoffman says she and her colleagues plan to expand the project to investigate more of the 100-plus known autism-linked genes.
