{"id":361427,"date":"2026-04-03T05:14:09","date_gmt":"2026-04-03T05:14:09","guid":{"rendered":"https:\/\/www.newsbeep.com\/nz\/361427\/"},"modified":"2026-04-03T05:14:09","modified_gmt":"2026-04-03T05:14:09","slug":"newly-identified-barrier-cells-seal-off-choroid-plexus-from-csf-rest-of-brain","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/nz\/361427\/","title":{"rendered":"Newly identified barrier cells seal off choroid plexus from CSF, rest of brain"},"content":{"rendered":"<p>A previously unrecognized population of fibroblasts seals off the base of the choroid plexus\u2014the network of blood vessels and cerebrospinal-fluid-producing epithelial cells that line the ventricles\u2014from the cerebrospinal fluid (CSF) and the rest of the brain, a new <a href=\"https:\/\/doi.org\/10.1038\/s41593-025-02188-7\" target=\"_blank\" rel=\"nofollow noopener\">study<\/a> in mice shows.\u00a0<\/p>\n<p>The newly identified barrier provides an added layer of protection that is distinct from the well-known blood-brain barrier and the one that the epithelial cells form between the blood and the CSF.\u00a0<\/p>\n<p>The findings help settle a long-standing debate about whether there was a blind spot in the choroid plexus that gave the periphery access into the brain, says <a href=\"https:\/\/www.tki.unibe.ch\/research\/research_groups\/britta_engelhardt\/index_eng.html\" target=\"_blank\" rel=\"nofollow noopener\">Britta Engelhardt<\/a>, professor of immunobiology at the University of Bern, who was not involved in the work. \u201cSome [scientists] speculated that there is a leak, like an opening, a secret window into the brain, and others said, \u2018No, there must be a barrier that we have overlooked.\u2019 And it\u2019s very obvious now.\u201d\u00a0<\/p>\n<p class=\"font-light italic text-2sm p-0 lg:text-3md\">\n            The study \u201chighlights how interconnected brain borders can be from very early in development.        <\/p>\n<p> \u2014 <\/p>\n<p>                 \u2014<br \/>\n                Carmen Ruiz de Almod\u00f3var <\/p>\n<p>Fibroblasts at the base of the choroid plexus, connected by adherens and tight junction proteins, cluster together around blood vessels and form a sealed barrier in mice, the researchers found. This structure represents a crucial component of compartmentalization in the choroid plexus, Engelhardt says. The cells were also present in human postmortem brain samples.\u00a0<\/p>\n<p>Similar to other barriers, the seal becomes leaky in response to inflammation triggered by lipopolysaccharide, a component of the bacterial cell wall, and it may coordinate immune cell crossing from the blood into the brain, the study also showed. The work was published in February in Nature Neuroscience.<\/p>\n<p>The results suggest that neuroscientists who study how immune cells interact with brain barriers may have been looking in the wrong place, says study investigator <a href=\"https:\/\/vandenbrouckelab.sites.vib.be\/en#\/\" target=\"_blank\" rel=\"nofollow noopener\">Roosmarijn Vandenbroucke<\/a>, professor of biomedical molecular biology at Ghent University. \u201cThat changes our focus a bit from not only looking at the blood-CSF barrier but also [\u2026] at the base barrier as a potential, maybe more preferred,\u201d point of entry for immune cells during inflammation.\u00a0<\/p>\n<p>And this novel structure can now be studied alongside other barriers, Engelhardt says. \u201cWe have now discovered that there is a connection from the outer leptomeninges all the way to the choroid plexus, and the question is: Where along this pathway are the barriers?\u201d she says. \u201cThis [study] is now setting the stage.\u201d<\/p>\n<p><p class=\"first-letter:text-2xl first-letter:float-left first-letter:text-red first-letter:pr-2 pb-0\">\n    V<\/p>\n<p>andenbroucke did not set out to find a new barrier. Rather, she was interested in using single-cell RNA sequencing to study the transcriptional variability of the choroid plexus in the different ventricles of the brain.\u00a0<\/p>\n<p>She failed to find any interesting signals, she says. \u201cTo be honest, the results were a bit disappointing.\u201d But when she looked more closely at the fibroblasts\u2019 expression, she noticed they segregated into two distinct populations. One of these showed barrier-like properties, including expression of the tight junction protein CLDN11, which piqued Vandenbroucke\u2019s interest.\u00a0<\/p>\n<p>These CLDN11-expressing fibroblasts resided at the base of the choroid plexus and clustered together, forming tight junctions that sealed the region, the researchers found through electron microscopy. The barrier was also functional, tracer experiments revealed. Tracer injected into the blood failed to access the CSF, whereas tracer injected into the ventricles remained there without crossing into the choroid plexus.<\/p>\n<p>The combination of these findings shows that \u201cit\u2019s a true, true barrier,\u201d Engelhardt says.\u00a0<\/p>\n<p>The fibroblasts, which the group named \u201cbase barrier cells,\u201d express many of the same genes as the cells that make up the meningeal arachnoid barrier, the study found. And they come from the meninges, lineage tracing shows, which \u201chighlights how interconnected brain borders can be from very early in development,\u201d says <a href=\"https:\/\/www.uni-bonn.de\/en\/research-and-teaching\/research-profile\/transdisciplinary-research-areas\/tra-3-life-and-health\/members-directory\/carmen-ruiz-de-almodovar\" target=\"_blank\" rel=\"nofollow noopener\">Carmen Ruiz de Almod\u00f3var<\/a>, professor of neurovascular cell biology at the University of Bonn, who was not involved with the study.\u00a0<\/p>\n<p><p class=\"first-letter:text-2xl first-letter:float-left first-letter:text-red first-letter:pr-2 pb-0\">\n    T<\/p>\n<p>he fact that fibroblasts created a seal was surprising, Engelhardt says. Many barriers, including the well-known blood-CSF barrier, are mainly composed of endothelial and epithelial cells, so neuroscientists did not have fibroblasts on their radar.
