{"id":571743,"date":"2026-08-17T14:00:12","date_gmt":"2026-08-17T14:00:12","guid":{"rendered":"https:\/\/www.newsbeep.com\/nz\/571743\/"},"modified":"2026-08-17T14:00:12","modified_gmt":"2026-08-17T14:00:12","slug":"human-heart-organoids-with-functioning-valves-model-cardiac-valve-disorders","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/nz\/571743\/","title":{"rendered":"Human heart organoids with functioning valves model cardiac valve disorders"},"content":{"rendered":"<p class=\"picture\"><img fetchpriority=\"high\" decoding=\"async\" alt=\"shutterstock_2801497411\" src=\"https:\/\/www.newsbeep.com\/nz\/wp-content\/uploads\/2026\/08\/22219_shutterstock_2801497411_897610.jpg\"   loading=\"eager\" class=\"lazyloaded\" width=\"2452\" height=\"1700\"\/><\/p>\n<p>Researchers have developed human heart organoids with functioning valves to model a range of heart valve disorders, creating a new platform for investigating disease mechanisms and potential treatments.<\/p>\n<p>The multidisciplinary team, led by Guang Li, an associate professor in<a href=\"https:\/\/www.cellbiology.pitt.edu\/\" target=\"_blank\" rel=\"noopener noreferrer nofollow\"> the University of Pittsburgh\u2019s School of Medicine Department of Cell Biology<\/a>, combined expertise in genetics, mechanics, chemistry and biology to develop the model.\u00a0<\/p>\n<p><a href=\"https:\/\/www.sciencedirect.com\/science\/article\/pii\/S1934590926002717?via%3Dihub\" target=\"_blank\" rel=\"noopener noreferrer nofollow\">The work<\/a> addresses a limitation of conventional animal models, which do not always reproduce human heart valve development and disease accurately.<\/p>\n<p>\u201chuman valves are very different from animal valves,\u201d Li said. \u201cTo study human valve diseases, we need human valve models.\u201d<\/p>\n<p>Building a human heart model<\/p>\n<p>The researchers used pluripotent adult human stem cells to create organoids, which are simplified versions of human organs. Different organoids can also be combined into assembloids to better reproduce the interactions between tissues found in complex organs.<\/p>\n<p>For the new model, Li\u2019s team grew a valve on the surface of a heart assembloid made by combining two organoids containing different types of heart cells.<\/p>\n<p>The researchers then incorporated mechanical forces designed to replicate conditions within a functioning heart. These included a flowing medium to mimic blood, an endothelial cell culture to model the cells lining heart valves and magnetised beads controlled by a magnetic belt to simulate muscle contraction.<\/p>\n<p>The work brought together researchers including Lance Davidson, William Kepler Whiteford Professor of Bioengineering in<a href=\"https:\/\/www.engineering.pitt.edu\/\" target=\"_blank\" rel=\"noopener noreferrer nofollow\"> the Swanson School of Engineering<\/a> and Si-Yang Zhen, Professor of Biomedical Engineering at <a href=\"https:\/\/www.cmu.edu\/\" target=\"_blank\" rel=\"noopener noreferrer nofollow\">Carnegie Mellon University.<\/a><\/p>\n<p>\u201cThis kind of project is really a hallmark of the community of researchers in Pittsburgh,\u201d Davidson said.<\/p>\n<p class=\"picture\"><img decoding=\"async\" alt=\"Low-Res_LiHeartAssembloid\" src=\"https:\/\/www.newsbeep.com\/nz\/wp-content\/uploads\/2026\/08\/22217_lowres_liheartassembloid_427333.jpg\"   loading=\"lazy\" class=\"lazyloaded\" width=\"700\" height=\"493\"\/><\/p>\n<p>Modelling heart valve disorders<\/p>\n<p>Once established, the organoid model was used to investigate four types of valve disorders, including mitral valve prolapse (MVP), a genetic condition affecting an estimated 7 to 8 million people in the US.<\/p>\n<p>The researchers introduced a mutation associated with MVP into the developing organoids. The resulting valves displayed characteristics of the disorder, allowing the team to investigate the pathways involved in its development.<\/p>\n<p>The researchers also created models of acquired valve damage, including changes associated with valve calcification, cryo-injury and complications linked to hypoglycaemia and diabetes.<\/p>\n<p>These models enabled Li and his colleagues to begin identifying biological pathways associated with valve abnormalities and investigate how they might be corrected. The team also plans to use the models to explore potential treatments for acquired valve disorders.<\/p>\n<p>Increasing model complexity<\/p>\n<p>The researchers aim to further develop the platform to more closely reproduce the structure and function of a human heart.<\/p>\n<p>Li\u2019s next step is to create assembloids containing two chambers and grow the valves inside them rather than on the surface.
