Yet much of their microbial genetic diversity remains unexplored, particularly in high-altitude ecosystems. A new study of the Napahai plateau wetland in Yunnan Province, China, has uncovered a distinctive pattern in DNA polymerase genes, suggesting that this isolated alpine wetland contains a genetic profile that differs from those found in marine, freshwater, hot spring, and agricultural environments.

“Our results suggest that the unusual geography and environmental conditions of the Napahai plateau wetland may leave a recognizable signature in the genes involved in DNA replication,” said corresponding author Xiuling Ji of Kunming University of Science and Technology. “These patterns give us a new way to examine how microbial communities differ across environments and how wetland ecosystems may influence genetic diversity.”

DNA polymerases are enzymes that copy DNA and help maintain genome stability. Because their genes vary among viruses, bacteria, archaea, and other organisms, researchers can also use them as molecular markers to investigate genetic diversity and evolutionary relationships.

The research team focused on three major DNA polymerase families, PolA, PolB, and PolC, using viral metagenomic data collected from soil and water in the Napahai wetland. The researchers compared Napahai sequences with DNA polymerase sequences from other environments and biological sources. In total, 1,222 DNA polymerase gene sequences were analyzed, including 104 from Napahai, 578 from other habitats, and 540 from other sources.

Phylogenetic analyses revealed that each polymerase family showed its own evolutionary structure. Importantly, Napahai sequences repeatedly formed cohesive and distinctive subclusters, even when they were compared with sequences from marine environments, lakes, hot springs, other wetlands, and paddy fields.

The same pattern appeared in ordination analyses used to visualize genetic similarities among environments. Principal coordinate analysis showed that Napahai DNA polymerase sequences occupied distributions that differed from those of several comparison habitats. Nonmetric multidimensional scaling also placed the Napahai sequences in a separate cluster, with a low stress value of 0.0546 and an R² value of 0.9739, indicating a strong representation of the observed distance patterns in the ordination.

The researchers suggest that Napahai’s unusual environmental setting may contribute to this differentiation. The wetland lies within the Three Parallel Rivers region near the Qinghai-Tibet Plateau and is influenced by distinctive alpine geography, seasonal hydrology, strong solar radiation, and climatic effects associated with Indian Ocean airflows. These factors may help shape the microbial communities and genetic variation found there.

The findings also raise the possibility that DNA polymerase genes could serve as biogeographical markers for distinguishing microbial communities from different habitats. However, the authors emphasize that the study is exploratory. Because the community-level analysis was based on two composite soil samples and one pooled water sample, the patterns require validation with greater biological replication.

Future studies across additional wetlands and terrestrial environments could determine whether similar DNA polymerase signatures occur elsewhere and clarify the ecological and evolutionary processes responsible for their distribution.

===

Journal reference: Shen X, Xiong L, Sun J, Wang T, Ji X. 2026.
