The ride will start at the BWFC Stadium and finish at Wembley, with the goal of raising funds for the Transforming Lives Group CIC, a mental health support organisation.
The team aims to complete the route in around 24 hours.
Andy Farnworth, founder of the Transforming Lives Group CIC, said: “This challenge is about more than the miles.
“It is about showing the people we support that we will go the distance for them.
“Mental health services are under enormous pressure and our project gives people something that cannot be prescribed, and that is community, connection and purpose.
“We hope the people of Bolton will get behind us.”
Mr Farnworth will be joined on the ride by Eddie Birchall, Darren Clare, Jason Grinnell and Robert Wysocki, with Paul Mockford as a support driver.
All funds raised will support the group’s Health and Wellbeing Support Project, which provides community-based activities designed to improve mental and physical wellbeing.
The project aims to reduce loneliness, promote recovery and offer a sense of belonging to some of the most vulnerable people in Bolton.
The team is calling on individuals and businesses across the town to support the challenge through donations or sponsorship.
