The creature is native to Mexico and has the ability to regrow its limbs (Picture: Tiia Monto\/Cover Media)<\/p>\n<p>A bizarre creature known for its enigmatic smile may hold the key to humans being able to regrow limbs, according to scientists.<\/p>\n<p>The axolotl is a critically endangered amphibian native to the lakes and wetlands south of <a data-ico=\"hyperlink-article\" href=\"https:\/\/metro.co.uk\/tag\/mexico\/\" data-track=\"inline-tag-auto-link_article\" rel=\"nofollow noopener\" target=\"_blank\">Mexico<\/a> City. <\/p>\n<p>Named after Xolotl, the Aztec god of fire, lightning and twins, it has been dubbed the Peter Pan of the salamander world as it stays in tadpole form forever and \u2018never grows up.\u2019<\/p>\n<p>But the reason scientists are so interested in it is because of its ability to regrow entire limbs and even parts of major organs.<\/p>\n<p>\t\t<img width=\"646\" height=\"430\" src=\"https:\/\/www.newsbeep.com\/uk\/wp-content\/uploads\/2026\/04\/coverimages55623953-f70a.jpg\" class=\"article-image wp-image-28044089\" alt=\"\" decoding=\"async\" loading=\"lazy\"\/><br \/>\n\t\tAn axolotl in Wake Forest University\u2019s regeneration lab, which is one of the institutions taking part in the study (PIcture: Wake Forest University\/Cover Media)<\/p>\n<p>The axolotl is being studied alongside mice and zebrafish, each chosen for its particular regenerative ability. <\/p>\n<p>Zebrafish can repeatedly regenerate their tail fins and several internal tissues. <\/p>\n<p>Mice, while far more limited, can regrow the tips of their digits \u2013 a capacity also seen in humans when the nail bed remains intact.<\/p>\n<p>In research published in the Proceedings of the National Academy of Sciences, the team say they have identified a common genetic mechanism that could one day support therapies to regrow human limbs.<\/p>\n<p>\u2018This significant research brought together three labs, working across three organisms to compare regeneration,\u2019 Josh Currie, assistant professor of biology at Wake Forest University, said. <\/p>\n<p>\u2018It showed us that there are universal, unifying genetic programs that are driving regeneration in very different types of organisms, salamanders, zebrafish, and mice.\u2019<\/p>\n<p>Globally, more than one million limb amputations take place each year due to conditions such as <a data-ico=\"hyperlink-article\" data-track=\"inline-tag-auto-link_article\" href=\"https:\/\/metro.co.uk\/tag\/diabetes\/\" rel=\"nofollow noopener\" target=\"_blank\">diabetes<\/a>, injury, <a data-ico=\"hyperlink-article\" href=\"https:\/\/metro.co.uk\/tag\/cancer\/\" data-track=\"inline-tag-auto-link_article\" rel=\"nofollow noopener\" target=\"_blank\">cancer<\/a>, and infection, according to Global Burden of Disease estimates. They focused on so-called SP genes, which they found to be essential for regeneration in all three species studied.<\/p>\n<p>Researchers found that regenerating skin in all three species activated two genes, SP6 and SP8. They then investigated how these genes function.<\/p>\n<p>Using gene-editing techniques, scientists removed SP8 from axolotls, preventing proper regrowth of limb bones. Similar effects were seen in mice lacking SP6 and SP8.<\/p>\n<p>\t\t<img width=\"646\" height=\"430\" src=\"https:\/\/www.newsbeep.com\/uk\/wp-content\/uploads\/2026\/04\/coverimages55623952-2662.jpg\" class=\"article-image wp-image-28044171\" alt=\"\" decoding=\"async\" loading=\"lazy\"\/><br \/>\n\t\tThe axolotl is known for its strange looks and a physiognomy which seems to be set in a permanent smile (Picture: Amanda Sofiarana\/Cover Images)<\/p>\n<p>Building on these findings, researchers developed an experimental gene therapy using a regeneration enhancer identified in zebrafish. The therapy delivered a molecule known as FGF8, helping to stimulate bone regrowth in mice and partially compensate for the missing genes.<\/p>\n<p>Although humans do not naturally possess this level of regenerative ability, the findings suggest it may be possible to replicate aspects of the process.<\/p>\n<p>Professor Currie, whose lab studies the Mexican axolotl, said: \u2018We can use this as a kind of proof of principle that we might be able to deliver therapies to substitute for this regenerative style of epidermis in regrowing tissue in humans.\u2019<\/p>\n<p>Researchers caution that significant <a data-ico=\"hyperlink-article\" href=\"https:\/\/metro.co.uk\/tag\/work\/\" data-track=\"inline-tag-auto-link_article\" rel=\"nofollow noopener\" target=\"_blank\">work<\/a> remains before such approaches could be applied to humans. However, they describe the study as an important step towards future treatments.<\/p>\n<p>\u2018Scientists are pursuing many solutions for replacing limbs, including bioengineered scaffolds and stem cell therapies,\u2019 Professor Currie added.<\/p>\n<p>\u2018The gene-therapy approach in this study is a new avenue that can complement and potentially augment what will surely be a multi-disciplinary solution to one day regenerate human limbs.\u2019<\/p>\n<p>He said that collaboration across different fields and model organisms was key to the breakthrough.<\/p>\n<p>\u2018Many times, scientists work in their silos: we\u2019re just working in axolotl, or we\u2019re just working in mouse, or just working in fish,\u2019 he explained. \u2018A real standout feature of this research is that we work across all these different organisms. That is really powerful, and it\u2019s something that I hope we\u2019ll see more of in the field.\u201d<\/p>\n<p>The study involved collaboration between scientists including David A. Brown, a plastic surgeon at Duke University, and Kenneth D.
