A new discovery of note in this field comes from experiments in <a href=\"https:\/\/en.wikipedia.org\/wiki\/Schizosaccharomyces_pombe\" rel=\"nofollow noopener\" target=\"_blank\">fission yeast<\/a> (an organism often used for studies of aging).<\/p>\n<p>Researchers from Queen Mary University of London have been testing a new drug called Rapalink-1, building on an existing immunosuppressant called rapamycin that has been shown to <a href=\"https:\/\/www.sciencealert.com\/scientists-confirm-anti-aging-drug-appears-to-prolong-life\" rel=\"nofollow noopener\" target=\"_blank\">extend the life of cells and rodents<\/a>. In these new tests, Rapalink-1 extended yeast lifespan to a similar degree as rapamycin.<\/p>\n<p>What&#8217;s more, molecular analysis revealed that the drug increased the production of enzymes that convert a compound made by gut bacteria, called <a href=\"https:\/\/en.wikipedia.org\/wiki\/Agmatine\" rel=\"nofollow noopener\" target=\"_blank\">agmatine<\/a>, into a variety of other chemicals.<\/p>\n<p>Related: <a href=\"https:\/\/www.sciencealert.com\/switching-off-one-crucial-protein-appears-to-reverse-brain-aging-in-mice\" rel=\"nofollow noopener\" target=\"_blank\">Switching Off One Crucial Protein Appears to Reverse Brain Aging in Mice<\/a><\/p>\n<p>Previous studies suggest agmatine can extend the lifespan of the microbes&#8217; host. Tests now reveal the conversion of the compound also indirectly affects genes linked to aging, which is part of the reason that Rapalink-1 was effective.<\/p>\n<p>While the science is quite technical, the takeaway is significant: It gives scientists a clearer indication of how yeast cells can be <a href=\"https:\/\/www.sciencealert.com\/eating-less-can-extend-lifespan-but-theres-a-hidden-catch-scientists-say\" rel=\"nofollow noopener\" target=\"_blank\">made to live longer<\/a>, through a method that may apply to human cells.<\/p>\n<p>&#8220;By showing that agmatinases are essential for healthy aging, we&#8217;ve uncovered a new layer of metabolic control over TOR \u2013 one that may be conserved in humans,&#8221; <a href=\"https:\/\/phys.org\/news\/2025-09-drug-enzyme-class-anti-aging.html\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> cellular biologist Charalampos Rallis.<\/p>\n<p>&#8220;Because agmatine is produced by diet and gut microbes, this work may help explain how nutrition and the microbiome influence aging.&#8221;<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/us\/wp-content\/uploads\/2025\/10\/YeastChart.jpg\" alt=\"Rapalink-1 effects chart\" width=\"642\" height=\"539\" class=\"wp-image-176110 size-full\"   loading=\"lazy\"\/>Rapalink-1 was shown to increase yeast cell longevity. Biol., 2025<\/a>)<\/p>\n<p>Both Rapalink-1 and rapamycin work through a biological pathway called the Target of Rapamycin, or TOR. This pathway is well known for its <a href=\"https:\/\/doi.org\/10.1016\/j.arr.2010.04.001\" rel=\"nofollow noopener\" target=\"_blank\">links to cell aging and disease<\/a>, first established in yeast, roundworms, flies, and mice. There&#8217;s a similar pathway <a href=\"https:\/\/doi.org\/10.1016\/S2666-7568(23)00258-1\" rel=\"nofollow noopener\" target=\"_blank\">in humans too<\/a>.<\/p>\n<p>Another important finding from the new study was that Rapalink-1 worked through TORC1, the part of the TOR pathway responsible for cell growth. Rapalink-1 puts the brakes on TORC1, which means cells <a href=\"https:\/\/www.sciencealert.com\/adults-grow-new-brain-cells-and-they-help-us-learn-through-listening\" rel=\"nofollow noopener\" target=\"_blank\">grow more slowly<\/a>, but last longer.<\/p>\n<p>&#8220;Understanding how TORC1 activity is tuned may be beneficial in both normal aging and also pathological states as well as in  <a href=\"https:\/\/www.sciencealert.com\/cancer\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73077\" data-postid=\"176106\" rel=\"nofollow noopener\" target=\"_self\">cancer<\/a> where TOR plays important roles,&#8221; the researchers <a href=\"https:\/\/dx.doi.org\/10.1038\/s42003-025-08731-3\" rel=\"nofollow noopener\" target=\"_blank\">write<\/a> in their paper.<\/p>\n<p>While you shouldn&#8217;t expect a miracle longevity tablet to arrive on the market anytime soon, these discoveries give us a better understanding of aging as an incredibly complex process that sees our bodies slowly break down and run out of steam <a href=\"https:\/\/www.sciencealert.com\/aging-can-spread-through-your-body-via-a-single-protein-study-finds\" rel=\"nofollow noopener\" target=\"_blank\">as the years go by<\/a>.<\/p>\n<p>That wearing down of our bodies increases the risk of a multitude of diseases, from  <a href=\"https:\/\/www.sciencealert.com\/go\/IaO\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73015\" data-postid=\"176106\" rel=\"nofollow noopener\" target=\"_self\">Alzheimer&#8217;s<\/a> to arthritis, and eventually, drugs like Rapalink-1 might keep some of those conditions at bay for longer. Rapalink-1 is already being trialed as a way of <a href=\"https:\/\/doi.org\/10.1038\/nature17963\" rel=\"nofollow noopener\" target=\"_blank\">fighting cancer tumors<\/a> and <a href=\"https:\/\/doi.org\/10.1016\/j.intimp.2023.111172\" rel=\"nofollow noopener\" target=\"_blank\">improving transplant outcomes<\/a>.<\/p>\n<p>However, the researchers emphasize that this work is still at an early stage, and that agmatine supplements \u2013 which are sold as promoting good health based on <a href=\"https:\/\/doi.org\/10.1089\/jmf.2014.0026\" rel=\"nofollow noopener\" target=\"_blank\">earlier studies<\/a> \u2013 should still be approached carefully.<\/p>\n<p>&#8220;We should be cautious about consuming agmatine for growth or longevity purposes,&#8221; <a href=\"https:\/\/phys.org\/news\/2025-09-drug-enzyme-class-anti-aging.html\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> Rallis. &#8220;Our data indicates the agmatine supplementation can be beneficial for growth only when certain metabolic pathways related to arginine breakdown are intact.&#8221;<\/p>\n<p>&#8220;In addition, agmatine does not always promote beneficial effects as it can contribute to certain pathologies.&#8221;<\/p>\n<p>The research has been published in <a href=\"https:\/\/dx.doi.org\/10.1038\/s42003-025-08731-3\" rel=\"nofollow noopener\" target=\"_blank\">Communications Biology<\/a>.<\/p>\n","protected":false},"excerpt":{"rendered":"Scientists continue to explore ways we can live longer and ensure those lives are healthier.
