{"id":342341,"date":"2025-12-11T04:20:16","date_gmt":"2025-12-11T04:20:16","guid":{"rendered":"https:\/\/www.newsbeep.com\/us\/342341\/"},"modified":"2025-12-11T04:20:16","modified_gmt":"2025-12-11T04:20:16","slug":"the-cause-of-alzheimers-may-be-coming-from-within-your-mouth-sciencealert","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/us\/342341\/","title":{"rendered":"The Cause of Alzheimer&#8217;s May Be Coming From Within Your Mouth : ScienceAlert"},"content":{"rendered":"<p>In recent years, an increasing number of scientific investigations have backed an alarming hypothesis:  <a href=\"https:\/\/www.sciencealert.com\/go\/IaO\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73015\" data-postid=\"185072\" rel=\"nofollow noopener\" target=\"_self\">Alzheimer&#8217;s<\/a> disease may not be merely a condition of an aging brain, <a href=\"https:\/\/www.sciencealert.com\/scientists-identify-a-virus-and-two-types-of-bacteria-that-could-be-linked-to-alzheimer-s\" rel=\"nofollow noopener\" target=\"_blank\">but the product of infection<\/a>.<\/p>\n<p>While the exact mechanisms of this infection are something researchers are <a href=\"https:\/\/www.sciencealert.com\/scientists-think-alzheimer-s-may-be-caused-by-our-brains-defence-system\" rel=\"nofollow noopener\" target=\"_blank\">still trying to isolate<\/a>, numerous studies suggest the <a href=\"https:\/\/www.sciencealert.com\/alzheimers-disease-beta-amyloid-source-outside-brain\" rel=\"nofollow noopener\" target=\"_blank\">deadly emergence<\/a> of Alzheimer&#8217;s <a href=\"https:\/\/www.sciencealert.com\/surgical-tools-could-spreading-alzheimers-proteins\" rel=\"nofollow noopener\" target=\"_blank\">goes way beyond<\/a> what we used to think.<\/p>\n<p>One such study, published in 2019, suggested what could be one of the most definitive leads yet for a bacterial culprit behind Alzheimer&#8217;s, and it comes from a somewhat unexpected quarter: gum disease.<\/p>\n<p>Related: <a href=\"https:\/\/www.sciencealert.com\/microbes-in-your-mouth-may-impact-your-risk-of-cognitive-decline\" rel=\"nofollow noopener\" target=\"_blank\">Microbes in Your Mouth May Impact Your Risk of Cognitive Decline<\/a><\/p>\n<p>Watch the video below for a summary of their study:<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/us\/wp-content\/uploads\/2025\/12\/1765426814_822_0.jpg\" alt=\"YouTube Thumbnail\" tabindex=\"0\" role=\"button\" class=\"youtube-thumbnail-preview\" loading=\"lazy\"\/> frameborder=&#8221;0\u2033 allow=&#8221;accelerometer; autoplay; clipboard-write; encrypted-media; gyroscope; picture-in-picture; web-share&#8221; referrerpolicy=&#8221;strict-origin-when-cross-origin&#8221; allowfullscreen&gt;<\/p>\n<p>In a paper led by senior author Jan Potempa, a microbiologist from the University of Louisville, researchers reported the discovery of <a href=\"https:\/\/en.wikipedia.org\/wiki\/Porphyromonas_gingivalis\" rel=\"nofollow noopener\" target=\"_blank\">Porphyromonas gingivalis<\/a> \u2013 the pathogen behind <a href=\"https:\/\/en.wikipedia.org\/wiki\/Chronic_periodontitis\" rel=\"nofollow noopener\" target=\"_blank\">chronic periodontitis<\/a> (aka gum disease) \u2013 in the brains of deceased Alzheimer&#8217;s patients.<\/p>\n<p>It wasn&#8217;t the first time the <a href=\"https:\/\/onlinelibrary.wiley.com\/doi\/full\/10.1111\/j.1532-5415.2010.02788.x\" rel=\"nofollow noopener\" target=\"_blank\">two factors have been linked<\/a>, but the researchers went further.<\/p>\n<p>In separate experiments with mice, oral infection with the pathogen led to brain colonization by the bacteria, together with increased production of amyloid beta (A\u03b2), the sticky proteins commonly associated with Alzheimer&#8217;s.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/newsletter?utm_source=promo_generic_health\" rel=\"nofollow noopener\" target=\"_blank\"><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/us\/wp-content\/uploads\/2025\/12\/Generic-Health-Promo-Final-642x273.jpg\" alt=\"Subscribe to ScienceAlert's free fact-checked newsletter\" width=\"642\" height=\"273\" class=\"alignnone wp-image-182810 size-medium\"   loading=\"lazy\"\/><\/a><\/p>\n<p>Related: <a href=\"https:\/\/www.sciencealert.com\/this-popular-diet-seems-to-reduce-gum-disease-scientists-say\" rel=\"nofollow noopener\" target=\"_blank\">This Popular Diet Seems to Reduce Gum Disease, Scientists Say<\/a><\/p>\n<p>The research team, coordinated by pharma startup Cortexyme, which was co-founded by first author Stephen Dominy, wasn&#8217;t claiming to have discovered definitive evidence of Alzheimer&#8217;s causation.<\/p>\n<p>But it was clear they thought we had a strong line of investigation here.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/us\/wp-content\/uploads\/2025\/12\/530-alzheimers-gum-disease-bacteria-1.jpg\" alt=\"530 alzheimers gum disease bacteria 1\" width=\"700\" style=\"width: 100%;\" height=\"429\" loading=\"lazy\"\/>P. gingivalis&#8217; (Cortexyme)<\/p>\n<p>&#8220;Infectious agents have been implicated in the development and progression of Alzheimer&#8217;s disease before, but the evidence of causation hasn&#8217;t been convincing,&#8221; <a href=\"https:\/\/www.medicalnewstoday.com\/articles\/324298\" rel=\"nofollow noopener\" target=\"_blank\">Dominy said at the time<\/a>.<\/p>\n<p>&#8220;Now, for the first time, we have solid evidence connecting the intracellular, Gram-negative pathogen, P. gingivalis, and Alzheimer&#8217;s pathogenesis.&#8221;<\/p>\n<p>In addition, the team identified toxic enzymes called gingipains secreted by the bacteria in the brains of Alzheimer&#8217;s patients, which correlated with two separate markers of the disease: the tau protein, and a protein tag called ubiquitin.<\/p>\n<p>Related: <a href=\"https:\/\/www.sciencealert.com\/stomach-ulcer-bacteria-could-be-a-surprise-ally-against-alzheimers\" rel=\"nofollow noopener\" target=\"_blank\">Stomach Ulcer Bacteria Could Be a Surprise Ally Against Alzheimer&#8217;s<\/a><\/p>\n<p>But even more compellingly, the team identified these toxic gingipains in the brains of deceased people who were never diagnosed with Alzheimer&#8217;s.<\/p>\n<p>That&#8217;s important, because while P. gingivalis and the disease have been linked before, it&#8217;s never been known \u2013 to put it simply \u2013 whether gum disease causes Alzheimer&#8217;s, or whether dementia leads to poor oral care.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/us\/wp-content\/uploads\/2025\/12\/dentist_gum_disease_642.jpg\" alt=\"dentist checking x rays\" width=\"642\" height=\"500\" class=\"wp-image-152675 size-full\"   loading=\"lazy\"\/>Does gum disease cause Alzheimer&#8217;s, or could those with dementia be at greater risk of poor dental hygiene? (<a href=\"https:\/\/unsplash.com\/photos\/person-in-gray-long-sleeve-shirt-holding-black-tablet-computer-v_2FRXEba94\" rel=\"nofollow noopener\" target=\"_blank\">Jonathan Borba\/Unsplash<\/a>)<\/p>\n<p>The fact that low levels of gingipains were evident even in people who were never diagnosed with Alzheimer&#8217;s could be a smoking gun \u2013 suggesting they might have developed the condition if they had lived longer.<\/p>\n<p>&#8220;Our identification of gingipain antigens in the brains of individuals with AD and also with AD pathology but no diagnosis of dementia argues that brain infection with P. gingivalis is not a result of poor dental care following the onset of dementia or a consequence of late-stage disease,&#8221; the authors explained <a href=\"http:\/\/advances.sciencemag.org\/content\/5\/1\/eaau3333\" rel=\"nofollow noopener\" target=\"_blank\">in their paper<\/a>.<\/p>\n<p>&#8220;But it is an early event that can explain the pathology found in middle-aged individuals before cognitive decline.&#8221;<\/p>\n<p>Further, a compound formulated by the company called COR388, showed in experiments with mice that it could reduce bacterial load of an established P. gingivalis brain infection, while also reducing amyloid-beta production and neuroinflammation.<\/p>\n<p>We&#8217;ll have to wait and see what future research will uncover about this link, but the research community is cautiously optimistic.<\/p>\n<p>Related: <a href=\"https:\/\/www.sciencealert.com\/we-may-now-know-why-alzheimers-erases-memories-of-our-loved-ones\" rel=\"nofollow noopener\" target=\"_blank\">We May Now Know Why Alzheimer&#8217;s Erases Memories of Our Loved Ones<\/a><\/p>\n<p>Today, Alzheimer&#8217;s disease is the most common form of dementia and a leading cause of death in the US.
