{"id":571773,"date":"2026-04-08T12:44:15","date_gmt":"2026-04-08T12:44:15","guid":{"rendered":"https:\/\/www.newsbeep.com\/us\/571773\/"},"modified":"2026-04-08T12:44:15","modified_gmt":"2026-04-08T12:44:15","slug":"new-drugs-take-aim-at-one-of-cancers-deadliest-mutations","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/us\/571773\/","title":{"rendered":"New drugs take aim at one of cancer\u2019s deadliest mutations"},"content":{"rendered":"<p> <img decoding=\"async\" class=\"figure__image\" alt=\"A light micrograph showing the pink and purple fibrous tissue of pancreatic cancer\" loading=\"lazy\" src=\"https:\/\/www.newsbeep.com\/us\/wp-content\/uploads\/2026\/04\/d41586-026-01016-7_52243880.jpg\"\/><\/p>\n<p class=\"figure__caption u-sans-serif\">A form of pancreatic cancer (pictured) is often caused by mutations in the KRAS protein.Credit: PJD\/Science Photo Library<\/p>\n<p>Armed with sophisticated chemistry and fresh approaches to drug design, researchers are closing in on innovative strategies to undermine <a href=\"https:\/\/www.nature.com\/articles\/d41586-022-03392-2\" data-track=\"click\" data-label=\"https:\/\/www.nature.com\/articles\/d41586-022-03392-2\" data-track-category=\"body text link\" rel=\"nofollow noopener\" target=\"_blank\">a cancer-causing mutant protein<\/a> once considered to be \u2018undruggable\u2019.<\/p>\n<p>Mutations in a protein called KRAS fuel some of the most lethal cancers. Designing drugs to disable KRAS has proved challenging \u2014 but this year, a field accustomed to disappointment has renewed hope. The first clinical trial of a drug that targets one form of mutant KRAS for degradation has shown signs of success<a href=\"#ref-CR1\" data-track=\"click\" data-action=\"anchor-link\" data-track-label=\"go to reference\" data-track-category=\"references\">1<\/a>. And four large clinical trials are testing another drug that inhibits several different mutant forms of KRAS and related proteins. The first of those studies is expected to report results in the next few months.<\/p>\n<p>It is unlikely that any of these approaches will, by themselves, provide a cure, says Dieter Saur, a gastroenterologist and cancer researcher at the Technical University of Munich in Germany. Instead, the hope is that they can be <a href=\"https:\/\/www.nature.com\/articles\/d41586-017-08720-5\" data-track=\"click\" data-label=\"https:\/\/www.nature.com\/articles\/d41586-017-08720-5\" data-track-category=\"body text link\" rel=\"nofollow noopener\" target=\"_blank\">combined with one another or with other drugs<\/a> to create a regimen that KRAS-mutant cancers cannot escape.<\/p>\n<p>\u201cIt\u2019s exciting. So many different things are going on,\u201d says Saur. \u201cThe field has completely changed.\u201d<\/p>\n<p>On too long<\/p>\n<p>KRAS belongs to the RAS family of proteins, which have key roles in cell growth and proliferation. Some KRAS mutations lock the protein in an \u2018on\u2019 position, leading to the uncontrolled cellular growth that is a hallmark of cancer.<\/p>\n<p><a href=\"https:\/\/www.nature.com\/articles\/d41586-022-03392-2\" class=\"u-link-inherit\" data-track=\"click\" data-track-label=\"recommended article\" rel=\"nofollow noopener\" target=\"_blank\"><img decoding=\"async\" class=\"recommended__image\" alt=\"\" src=\"https:\/\/www.newsbeep.com\/us\/wp-content\/uploads\/2026\/04\/d41586-026-01016-7_25512118.jpg\"\/><\/p>\n<p class=\"recommended__title u-serif\">Cancer drugs are closing in on some of the deadliest mutations<\/p>\n<p><\/a><\/p>\n<p>This made KRAS an appealing target for cancer drug development: find a drug that could switch it off again, the reasoning went, and it would no longer drive tumour growth. But the protein\u2019s relatively smooth surface posed a challenge to drug developers, who typically design molecules that inhibit a protein by binding to pockets in its surface.<\/p>\n<p>And when chemists finally found compounds that could bind to and disable mutant KRAS, the drugs turned out to be effective for only a short period of time. \u201cIn every patient, we see resistance developing,\u201d says Saur. \u201cIt\u2019s not a cure.\u201d<\/p>\n<p>That resistance came in a variety of forms, including new mutations in the KRAS protein, and activation of other cellular processes that could compensate for the loss of KRAS function<a href=\"#ref-CR2\" data-track=\"click\" data-action=\"anchor-link\" data-track-label=\"go to reference\" data-track-category=\"references\">2<\/a>. One hope was that some of this resistance could be avoided if, instead of inhibiting mutant KRAS protein, a drug was able to get rid of it altogether.<\/p>\n<p>Rubbish markers<\/p>\n<p>Enter the degraders, compounds that bind to KRAS and then tether it to another protein, called an E3 ubiquitin ligase.
