{"id":572184,"date":"2026-04-08T17:26:18","date_gmt":"2026-04-08T17:26:18","guid":{"rendered":"https:\/\/www.newsbeep.com\/us\/572184\/"},"modified":"2026-04-08T17:26:18","modified_gmt":"2026-04-08T17:26:18","slug":"alzheimers-risk-gene-alters-brain-activity-early-but-it-may-be-reversible-sciencealert","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/us\/572184\/","title":{"rendered":"Alzheimer&#8217;s Risk Gene Alters Brain Activity Early \u2013 But It May Be Reversible : ScienceAlert"},"content":{"rendered":"<p>Carrying one or two copies of the APOE4 gene variant significantly <a href=\"https:\/\/www.sciencealert.com\/alzheimers-gene-doesnt-guarantee-dementia-heres-what-you-can-do-to-reduce-risk\" rel=\"nofollow noopener\" target=\"_blank\">increases the risk<\/a> of developing  <a href=\"https:\/\/www.sciencealert.com\/go\/IaO\" class=\"lar_link lar_link_outgoing\" data-linkid=\"73015\" data-postid=\"197296\" rel=\"nofollow noopener\" target=\"_self\">Alzheimer&#8217;s<\/a>, and a new study reveals how APOE4 can change neuron activity \u2013 potentially <a href=\"https:\/\/www.sciencealert.com\/blood-test-clock-can-predict-alzheimers-symptoms-before-they-emerge\" rel=\"nofollow noopener\" target=\"_blank\">many years before symptoms<\/a> such as memory loss start showing.<\/p>\n<p>In young mice bred to have the APOE4 gene, researchers found specific neurons were smaller and more hyperactive in parts of the brain&#8217;s memory center, <a href=\"https:\/\/www.sciencealert.com\/researchers-discover-a-new-role-for-the-hippocampus-that-focuses-the-senses\" style=\"font-size: 18px; letter-spacing: -0.025em;\" rel=\"nofollow noopener\" target=\"_blank\">the hippocampus<\/a>.<\/p>\n<p>What&#8217;s more, they were able to identify a protein, Nell2, contributing to the disruption \u2013 and a potential pathway to <a href=\"https:\/\/www.sciencealert.com\/new-alzheimers-treatment-strategy-reverses-cognitive-decline-in-mice\" rel=\"nofollow noopener\" target=\"_blank\">reverse the damage<\/a> in advanced cases.<\/p>\n<p><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/us\/wp-content\/uploads\/2026\/04\/StudyResearcher.jpg\" alt=\"Dennis Tabuena at work\" width=\"642\" height=\"361\" class=\"wp-image-197299 size-full\"   loading=\"lazy\"\/>Neuroscientist Dennis Tabuena at work. (<a href=\"https:\/\/gladstone.org\/news\/how-alzheimers-risk-gene-disrupts-brain-circuits-long-memory-loss\" rel=\"nofollow noopener\" target=\"_blank\">Gladstone Institutes<\/a>)<\/p>\n<p>&#8220;To the best of our knowledge, this is the first study that has directly examined what APOE4 does to the function of neurons at different ages,&#8221; <a href=\"https:\/\/gladstone.org\/news\/how-alzheimers-risk-gene-disrupts-brain-circuits-long-memory-loss\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> senior author Misha Zilberter, neuroscientist at the Gladstone Institute of Neurological Disease in the US.<\/p>\n<p>&#8220;We found fundamental changes in brain circuits occurring in young mice that still had normal learning and memory, and importantly, that those changes predicted the development of cognitive deficits at older ages.&#8221;<\/p>\n<p>Previous human brain imaging studies have connected APOE4 to hippocampus hyperactivity in people with <a href=\"https:\/\/doi.org\/10.1523\/JNEUROSCI.4740-11.2011\" rel=\"nofollow noopener\" target=\"_blank\">mild cognitive impairment<\/a> or at <a href=\"https:\/\/doi.org\/10.1002\/ana.22105\" rel=\"nofollow noopener\" target=\"_blank\">risk of Alzheimer&#8217;s disease<\/a>. Here the researchers were able to scrutinize that relationship in greater detail than ever before, using animal models.<\/p>\n<p>Using a variety of analysis techniques, the team observed overexcited neurons in young APOE4 mice. The neurons in parts of the hippocampus in these animals had shrunk down, making them more excitable and easier to stimulate.<\/p>\n<p><a href=\"https:\/\/www.sciencealert.com\/newsletter?utm_source=promo_generic_health\" rel=\"nofollow noopener\" target=\"_blank\"><img decoding=\"async\" src=\"https:\/\/www.newsbeep.com\/us\/wp-content\/uploads\/2026\/03\/Generic-Health-Promo-Final-642x273.jpg\" alt=\"Subscribe to ScienceAlert's free fact-checked newsletter\" width=\"642\" height=\"273\" class=\"alignnone wp-image-182810 size-medium\"   loading=\"lazy\"\/><\/a><\/p>\n<p>The team also compared the neurons of APOE4 mice to those with <a href=\"https:\/\/www.sciencealert.com\/woman-who-defied-alzheimers-could-show-us-how-to-avoid-it\" rel=\"nofollow noopener\" target=\"_blank\">the APOE3 gene variant<\/a>, which carries a lower risk of developing Alzheimer&#8217;s.<\/p>\n<p>The researchers found that in the APOE3 mice, neurons also became more active \u2013 but not until old age. What&#8217;s more, the APOE3 mice didn&#8217;t develop cognitive problems like the APOE4 mice.<\/p>\n<p>Further investigation revealed the role of protein Nell2, which was abundant in the neurons of APOE4 mice. The researchers think this is the molecular mechanism through which APOE4 might make brains age before their time.<\/p>\n<p>When the researchers intervened to block Nell2 production in the mouse models, the neurons recovered: They returned to a normal size and went back to their regular patterns of firing activity. That&#8217;s a good sign <a href=\"https:\/\/www.sciencealert.com\/new-drug-stalls-alzheimers-development-in-breakthrough-trial\" rel=\"nofollow noopener\" target=\"_blank\">for potential treatments<\/a> further down the line.<\/p>\n<p>&#8220;What&#8217;s exciting about Nell2 is that we were able to reverse the disease manifestations in adult mice by lowering its level,&#8221; <a href=\"https:\/\/gladstone.org\/news\/how-alzheimers-risk-gene-disrupts-brain-circuits-long-memory-loss\" rel=\"nofollow noopener\" target=\"_blank\">says<\/a> Yadong Huang, neuroscientist at Gladstone.<\/p>\n<p>&#8220;That tells us the damage is not irreversible, and that there may be a window for intervention even after disease processes have been triggered.&#8221;<\/p>\n<p>These findings tie in nicely with previous Alzheimer&#8217;s research as well. High levels of the Nell2 protein <a href=\"https:\/\/doi.org\/10.18632\/oncotarget.24303\" rel=\"nofollow noopener\" target=\"_blank\">have been spotted<\/a> in the brains of Alzheimer&#8217;s patients before, for example, but this is the first time that it&#8217;s been connected to APOE4 in animal models.<\/p>\n<p>Alzheimer&#8217;s is a complex condition, <a href=\"https:\/\/www.sciencealert.com\/major-study-links-15-factors-to-early-dementia-risk\" rel=\"nofollow noopener\" target=\"_blank\">with multiple risk factors<\/a>, and there are major challenges in trying to identify how these risk factors influence each other, as well as how the causes driving Alzheimer&#8217;s progression differ from the <a href=\"https:\/\/www.sciencealert.com\/there-s-even-more-reason-now-to-stop-blaming-amyloid-plaques-for-alzheimer-s\" rel=\"nofollow noopener\" target=\"_blank\">consequences of the disease<\/a>.<\/p>\n<p>With APOE4 carriers accounting for up to <a href=\"https:\/\/doi.org\/10.1001\/jama.1997.03550160069041\" rel=\"nofollow noopener\" target=\"_blank\">three quarters<\/a> of Alzheimer&#8217;s cases, the <a href=\"https:\/\/www.sciencealert.com\/first-of-its-kind-map-of-alzheimers-reveals-hidden-gene-activity\" rel=\"nofollow noopener\" target=\"_blank\">genetic angle<\/a> is one that researchers are repeatedly returning to, in order to understand <a href=\"https:\/\/www.sciencealert.com\/new-unifying-theory-may-explain-how-alzheimers-emerges-in-the-brain\" rel=\"nofollow noopener\" target=\"_blank\">what exactly malfunctions<\/a> when the disease starts taking hold.<\/p>\n<p>Related: <a href=\"https:\/\/www.sciencealert.com\/insomnia-can-be-an-early-alzheimers-warning-sign-new-research-explains-why\" rel=\"nofollow noopener\" target=\"_blank\">Insomnia Can Be an Early Alzheimer&#8217;s Warning Sign.
