{"id":596540,"date":"2026-04-21T00:38:08","date_gmt":"2026-04-21T00:38:08","guid":{"rendered":"https:\/\/www.newsbeep.com\/us\/596540\/"},"modified":"2026-04-21T00:38:08","modified_gmt":"2026-04-21T00:38:08","slug":"new-treatment-lets-3-transplant-patients-halt-anti-rejection-drugs","status":"publish","type":"post","link":"https:\/\/www.newsbeep.com\/us\/596540\/","title":{"rendered":"New Treatment Lets 3 Transplant Patients Halt Anti-Rejection Drugs"},"content":{"rendered":"<p class=\"css-ac37hb evys1bk0\">Researchers at the University of Pittsburgh reported on Friday that they had trained the immune systems of a few patients to accept liver transplants without the drugs needed to avoid organ rejection.<\/p>\n<p class=\"css-ac37hb evys1bk0\">Three of eight patients have now been off the drugs for at least three years, perhaps an early step toward a new approach to transplantation that experts in the field have long hoped for. The <a class=\"css-yywogo\" href=\"https:\/\/doi.org\/10.1038\/s41467-026-71280-8\" title=\"\" rel=\"noopener noreferrer nofollow\" target=\"_blank\">study<\/a> was published in Nature Communications.<\/p>\n<p class=\"css-ac37hb evys1bk0\">\u201cThey\u2019ve gone a long way toward proving a principle,\u201d said Dr. William Jarnagin, a cancer surgeon at Memorial Sloan Kettering Cancer Center, who was not involved in the study.<\/p>\n<p class=\"css-ac37hb evys1bk0\">\u201cWith further development, it could really be a game changer,\u201d he added.<\/p>\n<p class=\"css-ac37hb evys1bk0\">Dr. Joohyun Kim, a liver transplant surgeon at Yale who was not involved with the study, said, \u201cEven 10 years ago, people thought \u2018tolerance induction\u2019 was a dream.\u201d<\/p>\n<p class=\"css-ac37hb evys1bk0\">Although anti-rejection drugs have made organ transplants possible, they come with grave consequences.<\/p>\n<p class=\"css-ac37hb evys1bk0\">The medications, which suppress the body\u2019s immune response, increase the risks of infection, cancer, high cholesterol levels, accelerated heart disease, diabetes and kidney failure. They can even cause hair loss.<\/p>\n<p>More on Organ Transplants<\/p>\n<p class=\"css-z9e7md\">An Improved System: A crackdown on problems with fairness and safety <a class=\"css-897g4k\" href=\"https:\/\/www.nytimes.com\/2026\/01\/23\/us\/organ-donation.html\" rel=\"nofollow noopener\" target=\"_blank\">is achieving results<\/a>, including a big drop in the number of sick patients being passed over for transplants.<\/p>\n<p class=\"css-z9e7md\">Skipped Over in Line: The sickest patients are supposed to get priority for lifesaving transplants. But more and more, <a class=\"css-897g4k\" href=\"https:\/\/www.nytimes.com\/interactive\/2025\/02\/26\/us\/organ-transplants-waiting-list-skipped-patients.html\" rel=\"nofollow noopener\" target=\"_blank\">they are being skipped<\/a>.<\/p>\n<p class=\"css-z9e7md\">Bungled Organ Retrievals:\u00a0Potential donors endured <a class=\"css-897g4k\" href=\"https:\/\/www.nytimes.com\/2025\/07\/20\/us\/organ-transplants-donors-alive.html\" rel=\"nofollow noopener\" target=\"_blank\">rushed or premature attempts<\/a>\u00a0to remove their organs. Some were gasping, crying or showing other signs of life.<\/p>\n<p class=\"css-z9e7md\">Courting Overseas Patients:\u00a0Despite an organ shortage, some U.S. hospitals <a class=\"css-897g4k\" href=\"https:\/\/www.nytimes.com\/2025\/12\/16\/us\/organ-transplants-international-patients.html\" rel=\"nofollow noopener\" target=\"_blank\">seek out international patients<\/a>, who pay as much as $2 million for a transplant. In recent years, patients from abroad have typically received organs faster than U.S. patients.<\/p>\n<p class=\"css-z9e7md\">Pig Organ Transplants: The U.S. Food and Drug Administration has given the green light to two biotechnology companies for clinical trials that will transplant organs from <a class=\"css-897g4k\" href=\"https:\/\/www.nytimes.com\/2025\/02\/03\/health\/pig-kidney-transplants-clinical-trials.html\" rel=\"nofollow noopener\" target=\"_blank\">genetically modified pigs into people with kidney failure<\/a>.<\/p>\n<p class=\"css-ac37hb evys1bk0\">Within five years of a liver transplant, 25 percent of patients, on average, have died. Many need kidney transplants because the anti-rejection drugs destroy their kidneys.<\/p>\n<p class=\"css-ac37hb evys1bk0\">\u201cThe list of adverse effects is long,\u201d said Dr. Abhinav Humar, chief of abdominal transplant services at the University of Pittsburgh Medical Center. \u201cIf you could avoid taking them, you would.\u201d<\/p>\n<p class=\"css-ac37hb evys1bk0\">Efforts to spare patients long-term dependence on anti-rejection drugs date back almost 30 years, said Dr. Angus Thomson, professor of surgery and immunology at the University of Pittsburgh.<\/p>\n<p class=\"css-ac37hb evys1bk0\">The research led to the idea that dendritic cells, a type of immune system cell, could be the key to preventing rejection. These cells help the body distinguish \u201cself\u201d from \u201cnon-self,\u201d an external threat.<\/p>\n<p class=\"css-ac37hb evys1bk0\">The idea, which became the basis of the new study, was that perhaps if an organ donor\u2019s dendritic cells were given to a recipient, the patient\u2019s body might learn to accept the new organ.<\/p>\n<p class=\"css-ac37hb evys1bk0\">The research team decided to start with liver transplants rather than the more common kidney transplants. Livers are perhaps the best tolerated transplanted organ.<\/p>\n<p class=\"css-ac37hb evys1bk0\">After taking immunosuppressive drugs for several years, 13 to 15 percent of liver transplant patients are able to stop. Their immune systems do not attack the transplanted livers.<\/p>\n<p class=\"css-ac37hb evys1bk0\">The investigators also elected to test their ideas with living donors. Because the liver can regenerate, doctors sometimes take a piece of a donor\u2019s healthy liver and transplant it. Both the donor and the recipient regrow full-sized livers.<\/p>\n<p class=\"css-ac37hb evys1bk0\">That allowed the researchers to time the surgery so they could give the recipient dendritic cells from the donor before the actual transplant. The hope was that the cells would prime the recipient\u2019s immune system to accept the new liver.<\/p>\n<p class=\"css-ac37hb evys1bk0\">The scientists said it might also be possible to use this method when a donor has died and there is no opportunity to wait before transplanting. Doctors would have to infuse the donor\u2019s dendritic cells a week after the transplant, because it takes a week to isolate and purify them.<\/p>\n<p class=\"css-ac37hb evys1bk0\">The <a class=\"css-yywogo\" href=\"https:\/\/www.nytimes.com\/2019\/01\/22\/health\/organ-transplants-immune-system.html\" title=\"\" rel=\"nofollow noopener\" target=\"_blank\">first patient<\/a> in the Pittsburgh study, Michael Schaffer, got his donor\u2019s dendritic cells and then a new liver in October 2017, but died suddenly and unexpectedly of unrelated causes eight years later. He was taking one anti-rejection drug, at the lowest dose, but was never able to quit it completely.<\/p>\n<p class=\"css-ac37hb evys1bk0\">One of the next patients, Barbara Bowser, did much better.<\/p>\n<p class=\"css-ac37hb evys1bk0\">Ms. Bowser, a 65-year-old retired school cleaner in Punxsutawney, Pa., had no idea her liver was failing. Then one day in December 2016, when she was 56, she felt sick to her stomach, went to the bathroom and passed out.<\/p>\n<p class=\"css-ac37hb evys1bk0\">\u201cThe bathroom looked like a crime scene,\u201d Ms. Bowser recalled. \u201cVarices in my esophagus had burst. My liver was not functioning.\u201d<\/p>\n<p class=\"css-ac37hb evys1bk0\">She wanted a living donor transplant, but no relatives were eligible. So, she said, \u201cI put my story on Facebook.\u201d A 33-year-old woman said she would donate.<\/p>\n<p class=\"css-ac37hb evys1bk0\">\u201cWe met on the day of the surgery,\u201d on Jan. 25, 2018, Ms. Bowser said. She had received an infusion of the donor\u2019s dendritic cells a week earlier.<\/p>\n<p class=\"css-ac37hb evys1bk0\">Ms. Bowser took anti-rejection drugs for nine months afterward and hated them \u2014 they made her feel ill, and her hair fell out. The Pittsburgh doctors were afraid to stop the medications immediately after a transplant.<\/p>\n<p class=\"css-ac37hb evys1bk0\">In October 2019, however, the Pittsburgh surgeons weaned Ms. Bowser off the drugs. She was terrified. \u201cI never thought it would work,\u201d she said.<\/p>\n<p class=\"css-ac37hb evys1bk0\">It has been six and a half years now.
