High-stakes human testing has now begun on a gene therapy that promises to reverse aging at the cellular level. About four months after the U.S. Food and Drug Administration (FDA) gave Life Biosciences clearance to <a href=\"https:\/\/www.wired.com\/story\/longevity-startup-doses-first-human-in-bid-to-reverse-age-related-sight-loss\/\" rel=\"nofollow noopener\" target=\"_blank\">initiate<\/a> its clinical trials, the Boston-based longevity startup announced on Tuesday that it\u2019s just dosed its first patient.<\/p>\n<p>Life Biosciences\u2019s modest goal is to regenerate aging neurons in the optic nerves of patients struggling with glaucoma\u2014a condition that often includes the irreversible death of neurons known as retinal ganglion cells, which connect the eye to the brain. The longevity firm\u2019s strategy involves injecting a modified virus with tailor-engineered genes that reprogram cells to \u201creverse age-related epigenetic changes,\u201d according to <a href=\"https:\/\/clinicaltrials.gov\/study\/NCT07290244\" rel=\"nofollow noopener\" target=\"_blank\">records<\/a> with the U.S. National Center for Biotechnology Information.<\/p>\n<p>But\u2014while the FDA has cleared the company\u2019s ER-100 drug for human tests\u2014some researchers watching this space worry that these \u201creprogramming\u201d therapies might lead to runaway (or exuberantly youthful) cellular growth, similar to past tests that may have caused <a href=\"https:\/\/www.technologyreview.com\/2019\/08\/08\/65461\/scientist-fountain-of-youth-epigenome\/\" rel=\"nofollow noopener\" target=\"_blank\">cancer<\/a> in mice.<\/p>\n<p>\u201cIt\u2019s gotten a lot of hype,\u201d neurobiologist Pete Williams at the Centre for Eye Research Australia <a href=\"https:\/\/www.nature.com\/articles\/d41586-026-01836-7\" rel=\"nofollow noopener\" target=\"_blank\">explained<\/a> to the journal Nature. \u201cIf this goes catastrophically wrong, it might screw us all in the future.\u201d<\/p>\n<p> Fresh eyes <\/p>\n<p>Life Biosciences focused on neurodegenerative diseases related to blindness, in part because the eye is a relatively isolated organ within the body where unintended consequences are less likely to get out of hand. To further increase its tests\u2019 safety, the company\u2019s ER-100 drug is only activated when patients simultaneously take the antibiotic doxycycline. Without it, the drug\u2019s genes switch off.<\/p>\n<p>\u201cWe\u2019re going to give doxycycline systemically,\u201d the company\u2019s chief scientific officer Sharon Rosenzweig-Lipson <a href=\"https:\/\/lifespan.io\/first-human-cellular-reprogramming-trial-cleared-by-the-fda\/\" rel=\"nofollow noopener\" target=\"_blank\">told<\/a> the nonprofit Lifespan Research Institute (LHI) this past February. The plan, she added, is to keep the genes activated over an eight-week period, more than the sporadic tests common in this field.<\/p>\n<p>\u201cWe can achieve good reprogramming and good safety with a more continuous expression system,\u201d Rosenzweig-Lipson said.<\/p>\n<p>The core of the company\u2019s treatment rests on breakthroughs by geneticist David Sinclair\u2019s <a href=\"https:\/\/sinclair.hms.harvard.edu\/\" rel=\"nofollow noopener\" target=\"_blank\">lab<\/a> at Harvard Medical School, which <a href=\"https:\/\/www.nature.com\/articles\/d41586-025-00032-3\" rel=\"nofollow noopener\" target=\"_blank\">showed<\/a> that activating three specific genes could regenerate neurons and reverse vision loss in both aging mice and mice with glaucoma. Since then, Life Biosciences\u2014which Sinclair co-founded\u2014says it has <a href=\"https:\/\/www.lifebiosciences.com\/life-biosciences-announces-fda-clearance-of-ind-application-for-er-100-in-optic-neuropathies\/\" rel=\"nofollow noopener\" target=\"_blank\">optimized<\/a> this approach via \u201ccomprehensive nonhuman primate studies.\u201d<\/p>\n<p>Still further beneath that foundational work, the company\u2019s gene therapy drug builds off the Nobel Prize-winning <a href=\"https:\/\/www.nobelprize.org\/prizes\/medicine\/2012\/yamanaka\/facts\/\" rel=\"nofollow noopener\" target=\"_blank\">discoveries<\/a> of stem cell researcher Shinya Yamanaka, who first <a href=\"https:\/\/biochem158.stanford.edu\/Stem%20Cell\/Yamanaka%20-%20Induction%20of%20stem%20cells.pdf\" rel=\"nofollow noopener\" target=\"_blank\">documented<\/a> the genes that can reprogram adult cells to return to a stem cell-like state.<\/p>\n<p>But, as you might imagine, it\u2019s the very promise of these genes as a \u201cfactory reset\u201d that can sometimes lead newly reprogrammed cells to veer off in the wrong direction. These \u201cYamanaka factor\u201d genes, as LHI noted, have produced <a href=\"https:\/\/lifespan.io\/topic\/yamanaka-factors\/#:~:text=This%20was%20the%20case,other%20partially%20formed%20organs\" rel=\"nofollow noopener\" target=\"_blank\">teratomas<\/a> in earlier experiments, \u201cbizarre tumors that are often found to contain hair, teeth, eyes, and other partially formed organs.\u201d<\/p>\n<p> The quest feel 10 years younger <\/p>\n<p>As Karl Pfleger, an investor in a U.K. competitor to Life Biosciences, <a href=\"https:\/\/www.technologyreview.com\/2026\/01\/27\/1131796\/the-first-human-test-of-a-rejuvenation-method-will-begin-shortly\/\" rel=\"nofollow noopener\" target=\"_blank\">told<\/a> MIT Technology Review this January, these early clinical trials are a long way from certain billionaire backers\u2019 <a href=\"https:\/\/www.nytimes.com\/2026\/04\/27\/magazine\/cell-rejuventation-biotech-longevity-research-altos-labs.html#:~:text=Fueling%20this%20industry,the%20aging%20process.\" rel=\"nofollow noopener\" target=\"_blank\">dreams<\/a> of <a href=\"https:\/\/www.msn.com\/en-us\/money\/companies\/the-billionaires-fueling-the-quest-for-longer-life\/ar-AA1M27h3\" rel=\"nofollow noopener\" target=\"_blank\">immortality<\/a>. But it\u2019s a start.<\/p>\n<p>\u201cThe optimistic case is this solves some blindness for certain people and catalyzes [future] work,\u201d Pfleger said. \u201cIt\u2019s not like your doctor will be writing a prescription for a pill that will rejuvenate you.\u201d<\/p>\n<p>That said, Sinclair is already gunning to do just that, <a href=\"https:\/\/www.technologyreview.com\/2026\/06\/09\/1138545\/david-sinclair-plans-to-test-whole-body-rejuvenation-drugs-in-the-xprize-competition\/\" rel=\"nofollow noopener\" target=\"_blank\">announcing<\/a> plans to develop an oral version of his genetic \u201creprogramming\u201d drugs as his bid to win a $101 million competition put on by the XPrize Foundation. The grand prize, ostensibly, will go to the team that can \u201crestore\u201d about 10 years to a patient\u2019s life, making them a decade younger in under one year of treatment.<\/p>\n<p>The contest will be something of a long shot. A recent attempt by one of Sinclair\u2019s XPrize competitors last year <a href=\"https:\/\/www.biorxiv.org\/content\/10.1101\/2025.06.25.661123v1\" rel=\"nofollow noopener\" target=\"_blank\">created<\/a> a toxic bloom of unwanted lipids in their test mice, \u201chindering the rejuvenation.\u201d<\/p>\n","protected":false},"excerpt":{"rendered":"Buckle up.
